摘 要:目的研究小窝蛋白-1(Caveolin-1)的表达与乙型肝炎病毒(HBV)感染以及相关肝细胞癌(HCC)的关系。方法免疫组化法检测HBV相关HCC中Caveolin-1表达量。构建Caveolin-1重组质粒,用Western blot检测转染了重组质粒的HepG2.2.15细胞中Caveolin-1表达量,并用ELISA法检测转染后培养上清中的HBsAg。结果免疫组化结果显示Caveolin-1在95%HBV相关HCC组织中表达低于癌旁组织(P<0.05)。Caveolin-1重组质粒构建成功并在HepG2.2.15中成功表达。ELISA检测发现用Caveolin-1重组质粒转染HBV感染稳定模型HepG2.2.15细胞48、72 h后,细胞培养上清中的HBsAg浓度相对未转染细胞分别下降了46.2%和48.8%(P<0.05)。重组质粒转染HBV感染急性细胞模型48、72 h后,HBsAg浓度相对未转染细胞分别下降了41%和50%(P<0.05)。结论 Caveolin-1在HBV相关HCC中呈低表达,Caveolin-1的高表达抑制了HBV HBsAg的分泌。Objective To detect the expression of Caveolin-1 in hepatitis B virus(HBV) related hepatocellular carcinoma(HCC) and determine the correlation between Caveolin-1 and HBV infection and HCC.Methods Immunohistochemical analyses were performed to detect the expression of Caveolin-1 in 40 tissue samples of HBV related HCC and 10 samples of non-related HCC,and their corresponding 50 paracarcinoma tissues.pcDNA3.1(+) Caveolin-1 eukaryotic expression vector was constructed and the recombinant plasmid was transiently transfected into HepG2.2.15 cells.The expression of pcDNA3.1(+) Caveolin-1 was detected by Western blotting.HBsAg expression of transfected cells were determined by ELISA.Results Immunohistochemical analyses results showed a lower expression of Caveolin-1 in 95% HBV-related HCC samples compared to their corresponding paracarcinoma samples(P0.05).Western blotting indicated that the recombinant plasmids was successfully transfected into HepG2.2.15 cells and Caveolin-1 was expressed.Using ELISA analysis we observed that the levels of HBsAg in the supernatants of transfected HepG2.2.15 cells were reduced by 46.2%(48 h,P0.05) and 48.8%(72 h,P0.05) compared with the control one.And also a lower secretion of HBsAg was found in the transfected HepG2.2.15 cells(decreased by 41% for 48 h and 50% for 72 h,when compared with the non-transfected cells,P0.05).Conclusion Caveolin-1 is expressed at a low level in HBV induced HCC,while the overexpression of Caveolin-1 might down-regulate HBV HBsAg secretion.
