摘 要:目的通过检测生存素、血管内皮生长因子C(VEGF-C)和增殖细胞核抗原(PCNA)在胆管癌中的表达水平,探讨其在胆管癌侵袭和转移中的作用机制。方法采用免疫组化过氧化物酶标记链霉卵白素法(SP)对42例胆管癌组织及20例正常组织中生存素、VEGF-C和PCNA进行检测。采用RT-PCR方法检测42例术中新鲜的胆管癌、17例同个体癌旁胆管黏膜和20例正常胆管组织中生存素、VEGF-C和PCNA的mRNA表达。结果生存素、VEGF-C和PCNA在胆管癌组织中表达的阳性率明显高于正常组织。胆管癌组织、癌旁组织、正常胆管组织中生存素mRNA相对表达量分别为0.624±0.080、0.569±0.044 vs 0.568±0.032(P<0.05);VEGF-C mRNA相对表达量分别为0.693±0.108、0.627±0.066 vs 0.611±0.058(P<0.01);PCNA mRNA相对表达量分别为0.630±0.077、0.569±0.041 vs 0.561±0.033(P<0.01);胆管癌组织、癌旁组织、正常胆管组织中生存素mRNA,VEGF-C mRNA和PCNA mRNA相对表达量在3组之间呈下降趋势(P<0.05)。结论生存素、VEGF-C、PCNA蛋白和mRNA在胆管癌组织中呈高表达,这些基因和蛋白表达的改变可能与胆管癌的发生及侵袭、转移等生物学行为密切相关。Objective To investigate the expression of survivin,vascular endothelial growth factor C(VEGF-C),proliferating cell nuclear antigen(PCNA) in cholangioncarcinoma and its relationship with the invasion and metastasis of cholangioncarcinoma.Methods Immunohistochemical technique SP was used to examine the expression of survivin,VEGF-C,PCNA in 42 cases of cholangioncarcinoma mucosa and 20 cases of normal tissues.The mRNA of survivin,VEGF-C,PCNA in 42 cases cholangioncarcinoma mucosa,17 cases paired adjacent mucosa to carcinoma and 20 cases of normal tissues were studied with RT-PCR.Results The positive expressions of survivin,VEGF-C,PCNA were all higher in cholangioncarcinoma mucosa than those of normal bile duct mucosa.The expression of survivin mRNA in cholangioncarcinoma mucosa,adjacent mucosa to carcinoma and normal bile duct mucosa was respectively 0.624±0.080,0.569±0.044 vs 0.568±0.032(P0.05);the expression of VEGF-C mRNA in cholangioncarcinoma mucosa,adjacent mucosa to carcinoma and normal bile duct mucosa respectively was 0.693±0.108,0.627±0.066 vs 0.611±0.058(P0.01);the expression of PCNA mRNA in cholangioncarcinoma mucosa,adjacent mucosa to carcinoma and normal bile duct mucosa was respectively 0.630±0.077,0.569±0.041 vs 0.561±0.033(P0.01);the expression of survivin mRNA,VEGF-C mRNA,PCNA mRNA was higher in cholangioncarcinoma than that of others(P0.05).Conclusion The expression of survivin,VEGF-C,PCNA protein and their mRNA in cholangioncarcinoma mucosa might have a close relationship with occurrence,invasion and metastasis of cholangioncarcinoma.
