人胰腺癌荷瘤裸鼠束缚应激模型的建立

摘　　要：目的建立人胰腺癌荷瘤裸鼠不良心理应激模型,以探讨不良心理应激对裸鼠移植瘤生长的影响。方法将20只裸鼠随机分为单纯荷瘤组和荷瘤+应激组,每组10只。荷瘤+应激组给予束缚应激。实验后测量两组裸鼠体质量、移植瘤质量与体积,采用酶联免疫吸附法(ELISA)检测两组裸鼠血清中肾上腺素(E)的含量。结果实验后荷瘤+应激组裸鼠体质量明显低于单纯荷瘤组(t=2.713,P<0.05);荷瘤+应激组瘤质量、瘤体积、血清E含量均高于单纯荷瘤组(t=-2.685～-2.486,P<0.05)。结论本实验采用束缚方法成功建立了人胰腺癌荷瘤裸鼠不良心理应激模型。Objective To create a distress psychological model in pancreatic-cancer-bearing nude mice and explore the influence of the distress on the growth of transplantation tumor in the model mice. Methods Twenty model mice were evenly randomized to two groups： pure-cancer-bearing group（PCB） and PCB plus stress（PCBPS） group.To those in PCBPS,shackle stress was offered.The body weight of the mice,the tumor weight and volume of transplantation tumors of both groups were measured,and serum epinephrine level detected with enzyme-linked immunosorbent assay（ELISA）. Results The average body weight of mice in the PCBPS was lower than that in the PCB（t=2.713,P0.05）;the weight and volume of the tumors,and serum epinephrine level were higher than that in the PCB（t=-2.685——2.486,P0.05）. Conclusion Using constraint method,a distress mental model in human pancreatic-cancer-bearing nude mice was successfully created.
