摘 要:目的探讨叶酸阿霉素纳米颗粒对体外培养的舌癌细胞超微结构的影响。方法体外培养的舌癌Tca8113细胞株分别采用阿霉素、叶酸纳米微粒、叶酸阿霉素纳米颗粒干预,透射电镜观察各组细胞超微结构变化。结果对照组细胞呈多角形,细胞内超微结构清晰;叶酸纳米微粒组细胞内内质网略减少;阿霉素组细胞失去多角形结构,线粒体肿胀,内质网脱颗粒,核糖体减少,可见少量凋亡小体;叶酸阿霉素纳米颗粒组细胞核膜崩解,细胞器消失,细胞核染色质固缩,细胞内可见较多凋亡小体。结论叶酸阿霉素纳米颗粒能够抑制口腔鳞癌细胞生长,引起舌癌细胞凋亡,其诱导细胞凋亡作用优于阿霉素单体。Objective To investigate the effect of folacin-adriamycin nanoparticles(FAN) on the ultrastructure of tongue cancer cells cultured in vitro. Methods The Tca8113 cell strains of tongue cancer cells cultured in vitro were intervened with adriamycin(ADM),folacin nanometer particles(FNP),and FAN,respectively.The ultrastructures of the cultured cells in the three groups were observed via transmission electron microscopy. Results The cells in the control group were polygonal in shape with clear ultrastructure;In FNP group,endoplasmic reticulum within the cells decreased;In ADM group,polygonal structure disappeared,mitochondria swelled,endoplasmic reticulum degranulated,ribosome decreased,and a small amounts of apoptotic bodies appeared;In FAN group,disintegration of the nuclear membrane,disappearance of the organelle,and more apoptotic bodies could be seen. Conclusion FAN can inhibit the growth of oral squamous cancer cells,leading to the apoptosis of tongue cancer cells.The induction of apoptosis of FAN is superior to adriamycin monomer.
