不同抗原制备大鼠实验性变态反应性脑脊髓炎的研究

摘　　要：目的：探索诱导ＳＤ大鼠发生实验性变态反应性脑脊髓炎（ＥＡＥ）的可能性，并寻找诱导ＳＤ大鼠ＥＡＥ的合适抗原，以推进国内临床神经免疫学的研究。方法：用含豚鼠、家兔、猪或牛脊髓的完全弗氏佐剂乳化物作为抗原，进行ＳＤ大鼠背部皮肤多点注射，观察和比较动物的神经症状和病理变化。结果：（１）各种抗原诱导ＥＡＥ，如果发病，一般都在注射抗原两周后开始出现症状，在注射后１９天左右症状最明显，且脑组织出现明显的炎性病理变化。（２）注射豚鼠脊髓抗原佐剂乳化物的大鼠发病率为１３％，注射牛脊髓的大鼠发病率为２５％，但注射豚鼠脊髓的大鼠ＥＡＥ症状重于注射牛脊髓的大鼠ＥＡＥ症状；注射兔或猪脊髓抗原佐剂乳化物的大鼠均未发病。结论：诱导ＳＤ大鼠发生ＥＡＥ可能以豚鼠和牛脊髓制备的抗原佐剂乳化物为好，但两者发病率都不高，可能与ＳＤ大鼠对抗原的敏感性较差等因素有关。Aim:Experimental allergic encephalomyelitis (EAE), a T-cell mediated autoimmune disease, is widely considered as an animal model of multiple sclerosis. Various antigens were used to induce EAE of Sprague-Dawley (SD) rats in order to search a fitter antigen that caused SD rats to get EAE. Methods: SD rats were injected in 5 to 6 locations of the back of the body 0.5 ml with the guinea pig, rabbit, pig or bull spinal cord in Freund's complete adjuvant and observed the clinical symptoms and histological alterations. Results: (1) The EAE clinical symptoms of SD rats occurred two weeks after the rats were injected with antigens. The peak of the clinical symptoms and histological alterations was on the 19th days. (2) The incidence of the disease injected with guinea pig spinal cord in Freund's complete adjuvant was 13% and injected with bull spinal cord in Freund's complete adjuvant was 25%. The EAE clinical symptoms injected with guinea pig spinal cord in Freund's complete adjuvant was more serious th antigenSprague-Dawley ratan with the bull spinal cord. The SD rats injected with rabbit or pig spinal cord in Freund's complete adjuvant all had not remarked clinical symptoms and histological alterations of EAE. TBXConclusion: guinea pig or bull spinal cord in Freund's complete adjuvant may be fitter antigens (for inducing EAE of SD rats.)
