Wistar大鼠胰岛素抵抗PCOS动物模型的构建及其特征分析

摘　　要：目的建立脱氢表雄酮(DHEA)诱导的大鼠多囊卵巢综合征(polycystic ovarian syndrome,PCOS)动物模型,并探讨其胰岛素抵抗(insulin resistance,IR)特征。方法将21日龄Wistar雌性大鼠随机分为PCOS组和对照组(每组20只),PCOS组每日皮下注射DHEA6 mg/100 g加0.2ml大豆油剂,连续20 d;对照组每日皮下注射0.2 ml大豆油剂,连续20 d。观察卵巢形态学改变,测定血中性激素、胰岛素样生长因子-1(IGF-1)、转化生长因子β1(TGFβ1)、空腹血糖(FPG)和空腹胰岛素(FINS)浓度,并计算胰岛素抵抗指数(HOMA1-IR)。结果 PCOS组卵巢囊性扩张卵泡和闭锁卵泡增多,发育卵泡减少;卵泡膜细胞增生。与对照组比较,PCOS组卵巢重量增加(P<0.05),血清睾酮(T)升高(P<0.01),FPG、FINS和HOMA1-IR均显著增高(P<0.05或P<0.01)。结论 DHEA可成功诱导Wistar大鼠胰岛素抵抗PCOS模型。Objective To establish the Wistar rat PCOS model with insulin resistance（IR）induced by dehydroepiandrosterone（DHEA）and to explore the characteristics of IR.Methods 22-day-old Wistar female rats were randomly divided into PCOS group and control group.The rats in the PCOS group were injected subcutaneously with DHEA dissolved in 0.2 ml soybean oil for 20 days,whereas the rats in the control group received injection of 0.2 ml soybean oil for 20 days.The ovarian morphology was observed using light microscopy（hematoxylin and eosin staining）.Serum levels of E2,T,LH,fasting blood glucose,serum insulin,insulin-like growth factor-1（IGF-1）and transforming growth factor β1（TGFβ1）were be detected and insulin resistance index（ISI）was calculated.Results When compared with the control group,PCOS group have more cystic dilatation of ovarian follicles and follicular atresia,while the number of follicles in the developmental stage and corpus luteum decreased significantly.Theca cell hyperplasia evidently in the PCOS group.The ovarian weight of the PCOS group was significantly higher than that in the control group（P0.05）.When compared with the control group,the serum T levels in the PCOS group were significantly higher（P0.01）,and FPG,FINS,and HOMA1-IR were significantly increased（P0.05 or P0.01）.Conclusion DHEA could successfully induce Wistar rat model of PCOS with IR.
