摘 要:以人工培养的发菜悬浮细胞为研究对象,通过分析低温胁迫下细胞的生长状态和生理特性的变化,阐释发菜在极端环境下得以生存的适应机制.结果表明:4C低温胁迫处理15天后,发菜悬浮细胞的相对生长量显著降低,而质膜相对透性和细胞活性在低温胁迫处理3天较对照有明显的变化,处理5天后细胞逐渐适应.与对照相比,低温胁迫处理5天使发菜细胞内的脯氨酸质量分数和海藻糖质量分数显著上升,同时显著抑制了光合系统Ⅱ的活性,而叶绿素a的质量分数在低温处理初期上升.由此可以发现,低温胁迫使发菜悬浮细胞的质膜完整性和光合作用受到伤害,而细胞内的脯氨酸和海藻糖迅速积累起着积极的渗透调节作用,从而使细胞逐渐适应低温,生长得以恢复.The artificial cultivation of Nostoc flagelliforme suspension cells was chose as the research object,in order to explain the adaptation mechanism of Nostoc flagelliforme for survive in extreme environments.Cell growth and physiological characteristics of Nostoc flagelliforme suspension cells under low-temperature stress were analyzed.The results showed that after 15 days under low temperature,the relative growth of Nostoc flagelliforme suspension cells decreased visibly.Meanwhile,the plasma membrane permeability and cell activity were apparently changed after 3 days under the low temperature.And then the cells gradually adapted to growth after 5 days.However,compared with control,intracellular proline content and trehalose content of Nostoc flagelliforme suspension cells increased significantly under the low temperature and the activity of photosystem Ⅱunder low temperature stress was inhibited distinctly,while the content of chlorophyll a increased at the low temperature at the beginning of the stress.The results indicate that under low-temperature stress,the physiological activity of Nostoc flagelliforme suspension cells could obviously be injuried,and the rapid accumulation of proline and trehalose played an active role in osmotic adjustment,so that the cells could gradually adapted themselves and grow at a low temperature.
