摘  要:采用实时荧光定量RT-PCR技术检测了条斑紫菜叶状体在25℃高温胁迫下PyHSP70-1基因的相对表达量变化。定量分析显示PyHSP70-1基因表达受到高温胁迫的显著诱导。在持续高温胁迫下,PyHSP70-1表达量在8 h时达到最大,此后一直维持在较高水平,且与对照差异显著。在短暂高温胁迫结束后PyHSP70-1表达量最大,随着恢复时间的增加,其mRNA量逐渐减少,在恢复的24 h时间内,mRNA量仍然显著高于对照。结果表明PyHSP70-1是诱导型HSP,受热激诱导表达,在热激胁迫中维持细胞蛋白与膜的稳定,对细胞具有重要的保护作用。Laver Porphyra yezoensis Ueda as one of the most economically important marine algae is extensively cultivated and consumed in eastern Asian.However,production and quality of the gametophytic blade are susceptible to high temperature.HSP70(70 kDa heat shock protein),one of the most conserved chaperones,plays a key role in cellular homeostasis under normal and stress.The expression profiles of HSP70 gene(designated as PyHSP70-1)was investigated in gametophytic blade of the laver under high temperature stress to explain,at least partially,its molecular mechanism of response to high temperature stress.The mRNA expression levels of PyHSP70-1 gene of the gametophytic blade were reported during and after heat shock at 25 ℃ using real-time RT-PCR.The maximal relative content of PyHSP70-1 mRNA was observed 8 hours after continuous high temperature stress,and then the high level maintained,significant different from that in the control.After short high temperature stress,the relative content of PyHSP70-1 mRNA reached the maximum level immediately,and then decreased gradually to a low level which was still significantly higher than that in the control during recovery phase of the first 24 h.These results showed that in PyHSP70-1,an inducible type HSP70,its expression was significantly induced by heat shock,and involved in protecting cells from heat shock.
