RRM2在人胆管癌组织及胆管癌细胞系中的表达和意义

摘　　要：目的:探讨核糖核苷酸还原酶M2(RRM2)在人胆管癌组织及胆管癌细胞系中的表达、分布以及生物学意义。方法:采用免疫组织化学技术(SP法)检测RRM2基因和蛋白在40例胆管癌标本、40例癌旁胆管组织以及10例正常胆管组织标本中的表达;同时采用RT-PCR法及Western blot法分别检测了RRM2基因mRNA及其蛋白在人胆管癌细胞系QBC939以及人正常胆管上皮细胞系HIBEC中的表达。结果:RRM2基因蛋白在胆管癌组织中表达阳性率为72.5%,而在癌旁胆管组织和正常胆管组织中未能检测到RRM2表达。在人胆管癌细胞系QBC939中,RRM2的mRNA及蛋白均特异性表达,而在正常人胆管上皮细胞系HIBEC中未见RRM2的表达。结论:RRM2在人类胆管癌组织和胆管癌细胞系中选择性高表达,可能与胆管癌的发生、发展密切相关。Objective：To study the expression,distribution and biological significance of ribonucleotide reductase M2 subunit（RRM2） in human cholangiocarcinoma tissues and QBC939 cell line.Methods：The expression of RRM2 was evaluated immunohistochemically in 40 cholangiocarcinoma samples,40 cases of adjacent histologically noncancerous bile duct,and 10 specimens of normal bile duct tissue.The expression of RRM2 in human cholangiocarcinoma cell line QBC939 and human normal bile duct epithelial cell line HIBEC were also detected by RT-PCR and Western blot analysis.Results：RRM2 was expressed in 72.5%（29 of 40） samples of carcinoma tissues,but it was undetectable in all adjacent histologically noncancerous bile and normal bile duct tissue.RRM2 was also expressed in cholangiocarcinoma cell line QBC939 but not in HIBEC.Conclusion：The high expression of RRM2 in human cholangiocarcinoma tissue and cell line suggest that RRM2 may be a key gene in initiation and progress of cholangiocarcinoma.
