摘  要:目的:研究中国小型猪痰瘀互结证冠心病模型中冠状动脉组织蛋白表达谱的改变情况。方法:采用冠状动脉球囊拉伤结合高脂饲料喂养建立中国小型猪痰瘀互结证冠心病模型,应用双向凝胶电泳、图谱分析软件、基质辅助激光解析-飞行时间质谱等蛋白质组学技术,对该模型动物的冠状动脉组织总蛋白进行双向电泳分离、差异比较、质谱鉴定及数据比对。:结果小型猪痰瘀互结证冠心病模型组冠状动脉组织与正常组相比共有蛋白差异点35个,14个上调点,17个特异表达点,另外模型组中有4个点发生未知修饰变化,累计鉴定出transgelin、apolipoprotein A-I等17个蛋白。结论:初步发现的差异蛋白可能与痰瘀互结证冠心病的形成、发生、发展相关,但是还需通过其它生物学方法做进一步验证。Objective : To explore the changes of Mini-pig coronary artery proteins expressing pattern related with syndrome of Phlegm and Blood Stasis(痰瘀互结证) of coronary heart disease.Methods : The model was established in mini-pig by high fat diet combined with balloon injury.The coronary artery total proteins of model were separated by two dimensional gel electrophoresis(2-DE).We compared protein expressing pattern in normal and model group by Image Master 2D Elite 6.01 analysis software.Then the different proteins were identified by MALDI-TOF-MS. Results: Compared with normal group,35 changed proteins had been determined: 14 proteins up-regulated,17 proteins specific expressed and 4 proteins unknow modified in mini-pig coronary artery tissure of the disease.With the search engine MASCOT,17 proteins including transgelin、apolipoprotein A-I etc,were identified.Conclusion : These proteins may play an important role in process of the formation,occurrence and development of the syndrome of phlegm and blood stasis of coronary heart disease.The result will be futher validated and investigated by other biological method.
