摘  要:目的观察雷公藤红素对高糖环境下大鼠视网膜血管内皮细胞VEGF和PEDF表达的影响。方法将体外培养的大鼠视网膜血管内皮细胞分为空白对照组、高糖组、高糖+雷公藤红素(0.05mg·L-1)组,进行相应处理后分别用免疫细胞化学及ELISA法检测细胞中VEGF和PEDF的表达。结果免疫细胞化学灰度值测定,空白组、高糖组及高糖+雷公藤红素组VEGF表达分别为74.40±4.19、110.12±7.32、78.11±5.81,PEDF表达量分别为124.65±6.01、88.73±7.84、93.31±5.67;ELISA结果显示空白组、高糖组及高糖+雷公藤红素组VEGF表达量分别为(155.21±13.36)ng·L-1、(237.69±19.41)ng·L-1、(168.57±26.32)ng·L-1,PEDF表达量分别为(251.37±17.95)ng·L-1、(178.35±25.36)ng·L-1、(191.58±31.37)ng·L-1,与正常对照组相比,高糖组VEGF表达增高(P<0.05),PEDF表达减少(P<0.05);与高糖组相比,高糖+雷公藤红素组VEGF表达减少(P<0.05),PEDF表达无明显差异性(P>0.05)。结论雷公藤红素能减少高糖环境下大鼠视网膜血管内皮细胞中VEGF表达,对PEDF表达的影响不明显。Objective To investigate the effect of tripterine on the expression of vascular endothelial growth factor(VEGF) and pigment epithelium derived factor(PEDF) of retinal vascular endothelial cells in mouse under high glucose condition. Methods Retinal vascular endothelial cells cultured in vitro were divided into blank control group,high glucose group and high glucose plus tripterine(0.05 mg·L-1) group.Expression of VEGF and PEDF were examined by immunocytochemistry and enzyme-linked immunosorbent assay. Results With immunocytochemistry,the expression of VEGF were 74.40±4.19,110.12±7.32,78.11±5.81,PEDF were 124.65±6.01,88.73±7.84,93.31±5.67 in blank control group,high glucose group and high glucose plus tripterine group;With enzyme-linked immunosorbent assay,VEGF were(155.21±13.36)ng·L-1,(237.69±19.41)ng·L-1,(168.57±26.32)ng·L-1,PEDF were(251.37±17.95)ng·L-1,(178.35±25.36)ng·L-1,(191.58±31.37)ng·L-1 in blank control group,high glucose group and high glucose plus tripterine group.Compared with normal control group,VEGF level significantly increased and PEDF level significantly decreased in high glucose group(both P〈0.05).Compared with high glucose group,VEGF level significantly decreased in high glucose plus tripterine group(P〈0.05),but PEDF level was without difference between the high glucose group and the high glucose plus tripterine group(P〉0.05). Conclusion Tripterine can decrease VEGF level in retinal vascular endothelial cells in mouse under high glucose,but it can not obviously effect the expression of PEDF.
