摘  要:目的:研究雷公藤内酯醇对阿尔茨海默病(AD)细胞模型海马神经元凋亡的影响,探讨雷公藤内酯醇治疗AD的可能机制。方法:用凝聚态Aβ1-40(20μg/ml)刺激的小胶质细胞条件培养液(MCM)作用于培养的大鼠海马神经元,建立AD细胞模型,应用MTT法和TUNEL染色,观察不同剂量的雷公藤内酯醇(5μg/ml和25μg/ml)在不同时程(2 h和24 h)内对AD细胞模型海马神经元凋亡的影响。结果:加药后2 h除模型MCM组海马神经元凋亡数高于正常对照组和正常MCM组(P<0.05)外,其余各组之间海马神经元凋亡数无明显差异。加药后24 h,模型MCM组海马神经元凋亡数较正常对照组和正常MCM组显著增多(P<0.01);低剂量用药MCM组和高剂量用药MCM组海马神经元凋亡数与模型MCM组比较显著降低(P<0.05,P<0.01);高剂量用药MCM组海马神经元凋亡数较低剂量用药MCM组明显降低(P<0.01)。结论:雷公藤内酯醇对AD细胞模型海马神经元的凋亡具有抑制作用。Objective:Effects of triptolide on neuronal apoptosis of hippocampus in AD cellular model were studied to explore the possible mechanism of triptolide against AD.Methods:The AD cellar model was established by action of microglial conditioned medium(MCM) induced by aggregated Aβ1-40(20 μg/ml) on rat cultured hippocampal neurons.MTT assay and TUNEL staining were used to observe the effects of triptolide at different dosages(5 μg/ml,25 μg/ml respectively) on neuronal apoptosis of hippocampus in AD cellar model in different phases(2 hours,24 hours respectively).Results:After 2 hours of cultivation,the apoptotic number of hippocampal neurons in model MCM group was more than that of normal control group and normal MCM group(P〈0.05),and the apoptotic number of hippocampal neurons in other groups have no significant difference.After 24 hours of cultivation,the apoptotic number of hippocampal neurons in model MCM group was obviously increased as compared with normal control group and normal MCM group(P〈0.01),and the apoptotic number of hippocampal neurons in low-dose triptolide MCM group and high-dose triptolide MCM group was obviously decreased as compared with model MCM group(P〈0.05,P〈0.01 respectively),and the apoptotic number of hippocampal neurons in high-dose triptolide MCM group was obviously decreased as compared with low-dose triptolide MCM group(P〈0.01).Conclusion:Triptolide can inhibit neuronal apoptosis of hippocampus in AD cellar model
