依达拉奉对大鼠血管性痴呆脑组织中Aβ、Tau和GFAP表达的影响

摘　　要：目的:研究自由基清除剂依达拉奉(edaravone)对血管性痴呆(vascular dementia,VD)大鼠皮质及海马区β-淀粉样蛋白(amyloidβ-protein,Aβ)、Tau蛋白和胶质纤维酸性蛋白(glial fibrillary acidic protein,GFAP)表达的影响,以探讨依达拉奉对脑的保护作用。方法:雄性SD大鼠48只,随机分为正常组、假手术组、VD病组和依达拉奉用药组,采用改良法建立SD大鼠VD模型。依达拉奉用药组于缺血后30 min腹腔内及皮下分别注射依达拉奉3 mg/kgwt,30 min后重复1次,以后每日1次。用免疫组织化学和免疫荧光法观察大鼠海马及皮质Aβ、Tau和GFAP免疫阳性细胞的变化。结果:依达拉奉用药组与VD病组相比较,大鼠海马及皮质Aβ、Tau和GFAP蛋白阳性细胞数明显减少(P<0.01或0.001)。Morris水迷宫(Morris Water Maze,MWM)测试显示,随着用药时间的延长,依达拉奉用药组与VD病组相比较,学习记忆有明显的改善(P<0.05)。结论:依达拉奉具有抑制脑内血管内皮细胞和神经细胞的过氧化作用,减少大鼠脑组织中由Aβ导致的神经细胞凋亡、降低脑组织中Tau和GFAP表达水平。Objective：To study the effects of free radical scavengers edaravone on the expressions of β-amyloid（Aβ）,Tau and glial fibrillary acidic protein（GFAP） in the cortex and hippocampus of vascular dementia（VD）.And to investigate the protective effects of edaravone on the brain.Methods：Male Spraque-Dawley（SD） rats（n=48）were randomly devided into 4 groups：normal group（n=6）,sham-operated group（n=6）,VD group（n=18） and edaravone-treated group（n=18）.We adopted improved method to make the VD model of SD rats.Edaravone-treated group rats were injected edaravone（3 mg/kg·wt） into peritoneal cavity and subcutaneous at 30 min after postischemia,repeat once after 30 min,and treated once every day later.The changes of the positive cellular numbers of Aβ、Tau and GFAP in the cortex and hippocampus were examined by immunohistochemical and immunofluorescent staining.Results：The positive cellular numbers of Aβ、Tau and GFAP in the cortex and hippocampus of the edaravone-treated group were distinctly less than those of the VD group（P〈0.001 or 0.01）.Morris Water Maze（MWM） test showed that with the prolong of medication,learning and memory ability in the edaravone-treated group was distinctly better than that in the VD group（P〈0.05 or 0.01）.Conclusion：Edaravone can inhibit peroxidation of vascular endothelial cell and neurons in the brain,reduce neuron apoptosis caused by Aβ in the rats brain,and decrease the protein level of Tau and GFAP in the brain
