摘 要:目的:研究九节龙皂苷Ⅰ(ardipusilloside Ⅰ)对口腔黏液表皮样癌Mc3细胞的增殖抑制与促细胞凋亡作用,以探讨九节龙皂苷Ⅰ的抗肿瘤作用机制。方法:利用MTT法检测九节龙皂苷Ⅰ对Mc3细胞生长抑制率的影响;采用FCM观察细胞周期并检测细胞凋亡率。结果:九节龙皂苷Ⅰ作用于Mc3细胞,在小剂量情况下对细胞杀伤作用较轻,随着药物浓度的增加,细胞生长抑制,其作用特点表现为明显的浓度依赖性,九节龙皂苷Ⅰ对Mc3细胞作用的IC50值为9.98μg/ml。10μg/ml和20μg/ml的九节龙皂苷Ⅰ组的凋亡率分别为24.83%±0.08%和42.63%±0.07%,与对照组7.63%±0.14%相比,凋亡率明显增高。结论:九节龙皂苷Ⅰ可抑制口腔黏液表皮样癌Mc3细胞的增殖并诱导细胞凋亡。Objective: To investigate the effect of ardipusilloside Ⅰ on the proliferation and apoptosis of mucoepidermoid carcinoma Mc3 cells.Methods: The inhibitory effect of ardipusilloside Ⅰ on Mc3 cells was measured by MTT assay.The apoptotic ratio and cell cycle analysis were determined by flow cytometry.Results:Ardipusilloside Ⅰ inhibited the proliferation of Mc3 cells in a dose-dependent manner.The IC50 value of ardipusilloside Ⅰ against Mc3 cells was 9.98 μg/ml.The apoptosis rates in 10 μg/ml and 20 μg/ml groups were 24.83%±0.08% and 42.63%±0.07% respectively,much higher than that in control group(7.63%±0.14%).Conclusion: Ardipusilloside Ⅰ can inhibit the proliferation and induce apoptosis of mucoepidermoid carcinoma Mc3 cells.
