摘  要:为了充分提高酶解效率及获得高活性的ACE抑制玉米肽,采用酶解与膜分离耦合技术连续化制备高活性ACE抑制玉米肽,考察了该制备过程中产品的质量和稳定性,比较了不同分子质量级分玉米肽的ACE抑制活性,以及采用HPLC-MS/MS对几种玉米肽的结构进行了解析。结果显示:在连续制备5 h内,所得产品的ACE抑制活性及肽含量较稳定,ACE抑制率在84%左右(最高达到89.82%),其中Mw<3 ku级分玉米肽其ACE抑制活性最高(IC50=0.29 mg/mL);解析出了3种玉米肽的结构,其氨基酸序列分别为QQLLPF、QQFLPF和QFLPF。试验表明:连续化酶膜耦合技术制备的ACE抑制玉米肽产品质量高且稳定,其中Mw<3 ku的玉米肽级分的ACE抑制活性最高;解析出的3种玉米肽均具有ACE抑制肽的结构特点。In order to improve enzymolysis efficiency and achieve high activity of angiotension I-converting enzyme(ACE) inhibitory corn peptides(CP),enzyme membrane coupling technology was used to prepare CP,and then ACE inhibitory activity of CP and the stability of quality were studied respectively.Besides,the primary structure of CP was analyzed by HPLC-MS/MS and the primary structure of some CP was confirmed by comparing corm proteins information in the MS database.The results showed that the ACE inhibitory activity and peptide level of hydrolysate were stable,their ACE-inhibitory activities were at approximately 84.00%(the highest was up to 89.82%) during 5 h of preparation by using enzyme membrane coupling technology.The fraction with MWCO3 ku exhibited the highest ACE inhibitory activity(IC50=0.29 mg/mL) than the other two fractions.The primary structures of three peptides were determined,including peptides 1∶QQLLPF,2∶QQFLPF,3∶QFLPF.The experiment exhibited that∶CP prepared by using enzyme membrane coupling technology had a stable and high quality,in which the fraction with MWCO3 ku exhibited the highest ACE inhibitory activity.The primary structures of three kinds of CP all had structural characteristics of ACE inhibitory peptide.
