摘 要:建立乳块消素片活性成分橙皮苷的空间分布和总体成分均匀性的质量控制方法,为片剂生物利用度与成分分布相关性研究提供基础。采用近/中红外漫反射光谱成像技术,运用基础相关系数分析方法,鉴别素片表面橙皮苷成分的分布区;通过直方图统计分析,探讨素片成分的分布均匀性。橙皮苷近红外光谱与素片表面光谱的最高相关系数为0.75;而中红外光谱与素片表面光谱的最高相关系数可达0.95以上。因此,中红外光谱成像更适用于乳块消素片橙皮苷空间分布的构建;直方图统计的标准差为0.05~0.09,峰度值为2.84~8.97,偏度值为-1.9~0.81,判定批次之间成分分布不均匀。研究表明,光谱成像技术可用于中药片剂成分的空间分布及分布均匀性研究。To provide the basis of research for the correlation between bioavailability and distribution of ingredients′,the quality control method of active ingredient Hesperidin distribution and composition uniformity of Rukuaixiao tablets intermediate was established.Using Near/Mid-Infrared diffuse reflectance spectral imaging technology,the distribution of the Hesperidin was identified based on correlation analysis method.Composition uniformity was explored by histogram analysis.Near infrared spectral imaging data,the highest correlation coefficient between pure Hesperidin spectra and the spectrum of surface composition was 0.75.As for infrared spectral imaging data,the correlation coefficient was above 0.95.The area was considered as the distribution of Hesperidin(Correlation coefficient 0.95).Furthermore,by histogram analysis of total absorption distribution,the range of value of standard deviation was from 0.05 to 0.09,kurtosis values,2.84—8.97,skewness value,-1.9—0.81.So the composition of tablets of different batches was believed as more unevenly.Therefore,the spectral imaging technique could be used to construct the spatial composition distribution and composition uniformity of the Traditional Chinese Medicine tablets.
