摘  要:对1株Ⅰ类新城疫病毒分离株NDV08-046进行了全基因组序列测定和遗传进化分析。结果表明,该分离株基因组长度为15198 nt,符合"六碱基原则"。与Ⅱ类新城疫病毒的基因组序列相比,该分离株在基因组P基因的2381~2382 nt的位置(根据La Sota株的基因组序列,包含15186 nt)有12 nt的插入(TGGGAGACGGGG)。NDV08-046株F蛋白裂解位点的序列为112-ERQERL-117,具有典型的新城疫弱毒株特征。全基因组的遗传进化分析显示,所有的新城疫毒株能够被分为2个明显不同的分支,即Ⅰ类和Ⅱ类,NDV08-046分离株在分类地位上属于Ⅰ类新城疫病毒基因2型。The complete genomic sequence of Newcastle disease virus(NDV) strain NDV08-046,isolated from domestic ducks in China,was determined in this study.The genome is 15 198 nucleotides(nt) in length,follows the "rule of six" and contains a 55-nt leader sequence at the 3′ end and a 114-nt trailer sequence at the 5′ end.Compared with the full genome sequences of class Ⅱ NDV strains,the NDV08-046 isolate has a 12-nt insertion(TGGGAGACGGGG) in the phosphoprotein gene between nucleotides 2 381 and 2 382 of the genome(numbered according to the genomic sequence of the La Sota strain,which consists of 15 186 nt).Strain NDV08-046 has the motif 112-ERQERL-117 at the cleavage site of the fusion protein,which is typical of lentogenic NDV strains.Phylogenetic analysis based on the full genome revealed that all the NDV strains studied could be divided into two distinct branches,namely class Ⅰ and class Ⅱ,and the NDV08-046 isolate characterized in this study was grouped in class Ⅰ.Further phylogenetic analysis based on a 374-bp fragment of the F gene in class Ⅰ strains of NDV demonstrated that NDV08-046 belongs to genotype 2.
