束缚应激致亚健康状态大鼠脑组织iNOS的表达及其中药干预作用

摘　　要：目的研究束缚应激致亚健康状态模型大鼠脑组织诱导型一氧化氮合成酶(iNOS)的表达情况,同时观察抗衰老片和六味地黄丸的干预作用。方法取雄性SD大鼠40只,随机分成4组,即正常对照组、模型对照组、抗衰老片组、六味地黄丸组,每组10只,采用每天限制活动3 h模拟人类心理应激,建立大鼠亚健康状态模型,造模同时,用抗衰老片、六味地黄丸进行干预。各组在造模第14 d、21 d分别取5只大鼠测定大脑皮质iNOS的表达。结果造模21 d后,模型对照组大鼠大脑皮质的iNOS表达显著增加(P<0.01);六味地黄丸可显著降低大鼠大脑皮质的iNOS阳性面积及总光密度(P<0.05)。结论束缚应激致亚健康状态大鼠大脑皮层的iNOS表达量增加,六味地黄丸可减少束缚应激致亚健康状态大鼠脑组织中iNOS的表达,预防和减少应激引起的神经元损伤,从而改善亚健康状态。Objective To study the expression of iNOS in brain tissue of sub-health state rat models induced by restraint stress,and observe the interventional effects of Anti-ageing Tablets and Liuwei Dihuang Pills.Methods 40 male SD rats were randomly divided into 4 groups： normal control group,model control group,Anti-ageing Tablets group and Liuwei Dihuang Pills group,10 rats per group.The sub-health state rat models were induced by means of movement limitation which simulated human physiological stress factor.At the same time,the rats of Anti-ageing Tablets group and Liuwei Dihuang Pills group were administrated by Anti-ageing Tablets and Liuwei Dihuang Pills.The expression of iNOS in brain tissue were detected on 14 and 21 days after moulding.Results 21 days after moulding,The level of iNOS in cerebral cortex of the rats of model control group was increased remarkably（P〈0.01）,while Liuwei Dihuang Pills could significantly reduce the expression of iNOS（P〈0.05）.Conclusion The expression of iNOS in cerebral cortex of the rats with the sub-health state induced by restraint stress is increase.Liuwei Dihuang Pills can reduce the expression of iNOS,prevent and reduce the neuron damage induced by restraint stress,thus improve the sub-health state.
