摘  要:目的 探讨理阿诺碱对雷帕霉素诱导的内皮生长晕细胞(EOCs)功能抑制的影响.方法 密度梯度离心法分离脐血单个核细胞,培养并扩增EOCs,免疫组化法、荧光染色法鉴定其内皮细胞特性.分别加雷帕霉素、理阿诺碱、理阿诺碱预处理1h再加雷帕霉素,与EOCs作用24h,CCK8检测细胞增殖能力,Transwell小室检测细胞迁移能力,并在倒置显微镜下检测细胞的黏附能力.结果 雷帕霉素抑制EOCs的增殖、迁移、黏附能力(P<0.05).理阿诺碱预处理1h能改善雷帕霉素对EOCs的增殖、迁移、黏附的抑制作用(P<0.05),理阿诺碱单独作用对EOCs的增殖、迁移、黏附无影响(P >0.05).结论 雷帕霉素抑制EOCs的增殖、迁移、黏附能力 理阿诺碱可以改善雷帕霉素诱导的对体外培养的EOCs增殖、迁移、黏附的抑制作用.Objective To investigate the effects of Ryanodine on Rapamycin-induced inhabition in endothelial outgrowth cells (EOCs). Methods The mononuclear cells were harvested from umbilical cord blood by Ficoll density gradient centrifugation, which are then induced into EOCs and expanded in vitro. The endothelial characteristics of EOCs were identified by immunostaining and fluorescent staining. The EOCs were pretreated with or without Ryanodine for lh, and then treated with or without Rapamycin for 24h. Cell proliferation was evaluated by CCK8 and migration was examined by Transwell. Adhesion of E^Cs was measured under the inverted microscopy by counting adherent cells after ceil replating. Results Rapamycin down-regulated proliferation, migration and adhesion abilities of EOCs (P〈0.05). Pretreated with Ryanodine enhanced the inhibitive effects on EOCs proliferation, migration and adhesion induced by Rapamycin (P〈0.05). Treated with Ryanodine alone did not affect proliferative, migratory, adhesive ability of EOCs compared to control group (P 〉0.05). Conclusion Ryanodine can enhance rapamycin-induced inhibitive effects on proliferative, migratory, adhesive abilities of EOCs.
