急性肝衰竭大鼠肝匀浆蛋白诱导骨髓间充质干细胞向肝细胞的分化

摘　　要：目的研究D-氨基半乳糖/内毒素介导的急性肝衰竭大鼠肝匀浆蛋白对骨髓间充质干细胞(BMSCs)分化的影响。方法将D-氨基半乳糖/内毒素腹腔注射制备而成的肝衰竭大鼠肝匀浆蛋白加入培养液中培养BMSCs,通过观察BMSCs形态以及检测ALB、AFP、糖原、尿素氮的合成来观察它对细胞分化的影响。结果肝衰竭大鼠肝匀浆按分子量的大小可以分为6个部分,其中4个部分的蛋白可以导致BMSCs转变成较为典型的多角型、卵圆形细胞,并且这些细胞中的AFP、ALB、糖原染色、培养液中尿素氮浓度也有明显增加,以第3、第4部分蛋白更为显著。结论急性肝衰竭模型肝匀浆蛋白可以促进BMSCs向肝细胞样细胞分化,其中以第3、第4波峰蛋白更为显著。【Objective】 To investigate the effects of liver failure rat liver Homogenate on rat BMSCs s differentiation into hepatocyte-like cells.【Methods】Rat BMSCs were cultured for the study.Acute liver fail-ure was induced in male Sprague-Dawley rats by intraperitoneal injection of galactosamine（D-GalN） and Lipopolysaccharides（LPS）.BMSCs were treated by isolated and purified protein from acute liver failure rat liver Homogenate,then the early hepatocyte markers alpha-fetoprotein（AFP）,the late liver-specific markers albumin（ALB） and hepatic speciial functions including ALB secretion,glycogen storage,urea production were detected.【Results】Protiens in liver failure rat liver Homogenate were isolated into six fractions according to the size of molecular weight.Upon been exposured to the four prior fractions,expression of AFP was rapidly detected,level of ALB（gene） was gradually upregulated.In addition,glycogen storage and urea production can be observed earlier.【Conclusion】Isolated and purified protein from acute liver failure rat liver Ho-mogenate can induce BMSCs to differentiae into hepatocyte-like cells rapidly and the third and fourth frac-tions can do that more efficiently.
