摘 要:目的:首次采用提取溶媒中加盐酸的方法测定升麻药材中阿魏酸、异阿魏酸和咖啡酸的含量。方法:采用Hypersil BDS C18(200mm×4.6mm,5μm)色谱柱,乙腈-0.1%磷酸水(15∶85)为流动相,测定波长为316nm,流速为1.0mL/min,柱温为30℃。结果:阿魏酸、异阿魏酸和咖啡酸分别在0.312~2.808mg(r=0.9998,n=5)、0.270~2.430mg(r=0.9995,n=5)和0.234~2.106mg(r=0.9997,n=5)线性关系良好,精密度、稳定性、重现性和加样回收率试验的RSD均小于3%,平均回收率分别为99.29%、99.73%和99.43%。结论:该方法准确、快速、重现性好,可用于同时测定升麻药材中阿魏酸、异阿魏酸和咖啡酸的含量。通过对升麻3个种不同产地中阿魏酸、异阿魏酸和咖啡酸含量的测定,为合理评价升麻药材质量奠定了基础,也为升麻药材质量的控制方法提供了一种新的参考依据。Objective:To establish a method for the determination of ferulic acid,isoferulic acid and caffeic acid in the pulp of rhizoma Cimicifugal by RP - HPLC simultaneously, which was added HC1 in extraction solvent firstly. Methods:The ferulic acid, isoferulie acid and caffeie acid were separated on Hypersil BDS C18 eoloum(200mm×4. 6mm,5m). Acetonitrile-0. 1% phos-phoric acid (15:85) was used as mobile phase , the detection wavelength was316nm, the flow rate was 1. 0mL/min and the column temperature was30℃. Results : The linear range of ferulic acid, isoferulic acid and caffeic acid Separately were within 0. 312 - 2. 808mg( r = 0. 9998, n = 5 ) ,0. 270 - 2. 430mg( r = 0. 9995, n = 5 ) and 0. 234 - 2. 106mg( r = 0. 9997, n = 5 ). The results that RSD of precision,stability ,reproducibihty,sample recovery and processing tests were all less than3%. The average recoveries of ferulic acid,isoferulic acid and caffeic acid were respectively 99. 29% ,99.73% and 99.43%. Conclusion:The method is quick, accurate and reproducible. It can be used for the determination of ferulic acid,isoferulic acid and caffeic acid in the pulp of rhizoma Cimicifugal simultaneously. This also provides a new frame of reference and lays a foundation for the reasonable evaluation of the quality of cimicifugal and the quality control through the determination of ferulic acid, isoferulic acid and caffeic acid in different regions which all come from three species of rhizoma Cimicifugal.
