摘 要:目的:研究乳腺癌中microRNA-21(miR-21)的表达及其与转化生长因子-β1(TGF-β1)的关系。方法:用不同质量浓度的TGF-β1 0、1.25、2.5、5、10 ng/mL干预MCF-7细胞24 h,以及不同时间(0、6、12、24、48、72 h)TGF-β1干预MCF-7细胞,用茎环实时荧光定量聚合酶链反应法(Real-time PCR)测定miR-21的表达。分别用实时定量PCR和免疫组织化学SP法检测37例乳腺癌中miR-21和TGF-β1的表达情况及其相关性。结果:茎环实时定量PCR法检测miR-21表达的敏感性和特异性良好,MCF-7细胞中miR-21的表达随TGF-β1作用呈时间依赖性和浓度依赖性。乳腺癌组织中miR-21的表达与淋巴结转移及TGF-β1的表达程度有关(P<0.05)。结论:高表达的TGF-β1可引起miR-21表达升高,与乳腺癌的侵袭转移有密切关系。Aim:To investigate the expression of microRNA-21(miR-21)and transforming growth facor-β1(TGF-β1) and to explore their relationship in breast cancer.Methods: MCF-7 was treated with different concentrations of TGF-β1(0、1.25、2.5、5、10 ng/ mL)for 24 h or treated with 10ng/ml TGF-β1 at different time(0、6、12、24、48、72 h),the expression of miR-21 was detected by stem-loop real-time PCR.In thirty-seven breast cancer cases,to explore the relationship of miR-21 and TGF-β1,the expression of miR-21 and TGF-β1 were detected by real-time PCR or immunohistochemical staining,respectively.Results: The sensitivity and specificity of stem-loop real-time PCR allowed for direct analysis of expression of miR-21 in breast cancer cell line and tissues.The expression of miR-21 was time-dependent and concentration-dependent after treated with TGF-β1 in MCF-7.The expression of miR-21 was correlated with lymph node metastasis and the expression of TGF-β1.Conclusion:High level of expression of miR-21 are correlated with infiltration and metastasis of breast cancer,and it is also correlated with high expression of TGF-β1.
