益肾蠲湿合剂对TGF-β_1诱导的肾小球系膜细胞分泌细胞外基质的影响

摘　　要：目的探讨中药复方益肾蠲湿合剂对TGF-β1作用下肾小球系膜细胞(GMC)分泌细胞外基质的影响。方法将含不同剂量益肾蠲湿合剂的药物血清与TGF-β1(终浓度5μg/L)共同培养GMC 24,48,72 h,收集培养上清并用ELISA法检测纤维连结蛋白(FN)、层黏连蛋白(LN)及Ⅳ型胶原(ColⅣ)含量。结果益肾蠲湿合剂的药物血清可抑制TGF-β1刺激GMC分泌FN、LN和ColIV,而且随着药物浓度的增加,其对GMC分泌FN、LN和ColIV的抑制率也相应增加。结论益肾蠲湿合剂对TGF-β1诱导的GMC分泌FN、LN和ColIV有明显的抑制作用,提示抑制TGF-β1刺激GMC分泌细胞外基质是本复方治疗慢性肾小球疾病的重要机制之一。Objective It is to explore the effect of traditional Chinese herbs Yishenjuanshi mixtures on the secretion of extracellular matrix induced by TGF-β1 in cultured glomerular mesangial cells（GMCs）.Methods GMCs were cultured for 24,28,72 h by TGF-β1（final concentration 5 μg/L） and drug serum of different dosage of Yishenjuanshi mixtures.The supernatant was collected respectively to determine the concentration of fibronectin（FN）,laminin（LN） and collagen Ⅳ（ColⅣ） by ELISA.Results Yishenjuanshi mixtures significantly inhibited GMC to secrete FN,LN and ColⅣ.The inhibition ratios increased with the dose of Yishenjuanshi increase.Conclusion Yishenjuanshi mixtures can inhibit cultured GMC to secrete FN,LN and ColⅣwhich induced by TGF-β1,it indicates that inhibiting TGF-β1 to stimulate GMC to secrete extracellular matrix is one of the important mechanisms of this medicine for treating for chronic renal glomerular disease.
