八聚体结合蛋白4在不同转移能力肿瘤细胞株中的表达

摘　　要：目的研究不同转移能力肿瘤细胞株中八聚体结合蛋白4(Oct 4)的表达情况及其意义。方法选取3对不同转移能力的肿瘤细胞株:高转移能力人乳腺癌细胞MDA-MB231和低转移能力人乳腺癌细胞MCF7,高转移能力人食管癌细胞Eca-109和低转移能力人胃腺癌细胞AGS,高转移能力卵巢癌细胞HO-8910PM和低转移能力人卵巢癌细胞SK-OV-3;采用RT-PCR、实时荧光定量PCR和细胞免疫荧光技术分别从mRNA和蛋白水平检测Oct 4在各株肿瘤细胞中的表达情况。结果 Oct 4在6株肿瘤细胞中mRNA和蛋白水平均有表达。高转移能力细胞中mRNA表达量较低转移能力细胞高;Oct 4蛋白红色荧光在细胞质中均有显示,在高转移能力细胞中较低转移能力细胞亮。结论 Oct 4在高转移能力肿瘤细胞株中的表达较低转移能力肿瘤细胞高,提示与肿瘤细胞的转移能力可能存在相关性,其机制有待进一步研究。Objective To investigate the expression of transcription factor Oct 4（octamer-binding transcription factor 4 ） in spontaneous metastatic potential of tumor cells,and discuss the investigation significance. Methods Three pairs of cells,with different spontaneous metastatic potentia,were collected, they were human breast cancer cell MDA-MB231 and MCF7,human esophageal cancer Eca-109 and gastric epithelial cell AGS,high-metastatic human OVCA cell line HO-8910PM and ovarian cancer cell SKOV -3.The ability of metastatic of the former was stronger than that of the latter.Through reverse transcription PCR（RT-PCR）,real-time PCR and immunocytochemistry methods,the mRNA levels and protein expression of Oct 4 were detected in tumor cells.Results Both mRNA and protein level,the positive expression of Oct 4 were detected in six cells.In addition,the mRNA expression level of Oct 4 in high-metastatic cells was higher than that in the low-metastatic cells.As far protein level,Oct 4 expression in high-metastatic cells was obviously lighter than that in the low-metastatic cells.Conclusion The relationship between the expression levels of Oct 4 and the metastatic of tumor cells may be correlative.The function of transcription factor Oct 4 in tumor cells metastatic mechanism is to be confirmed.
