摘  要:目的探讨熊果酸(UA)对人肝癌SMMC-7721细胞周期的影响。方法用不同浓度的UA处理SMMC-7721细胞,采用四甲基偶氮唑盐(MTT)法测定细胞增殖的抑制作用,流式细胞仪检测细胞周期的变化,RT-PCR检测细胞周期相关基因p21、p53及增殖细胞核抗原(PCNA)的表达。结果 UA对SMMC-7721细胞生长有显著的抑制作用,且此作用呈明显的时间、剂量依赖性;UA可以诱导SMMC-7721细胞阻滞在S期,RT-PCR结果显示其能够上调p21和p53基因,下调PCNA基因。结论 UA能明显抑制SMMC-7721细胞的生长,引起细胞周期阻滞;细胞周期阻滞的发生与p21、p53、PCNA基因变化相关。Objective To investigate effect of ursolic acid on cell cycle regulation of human hepatoma cell line SMMC-7721.Methods After treatment with UA,the growth inhibition of SMMC-7721 cells was assessed by MTT colorimetric assay.The effect of UA on the cell cycle phase distribution of SMMC- 7721 cells was analyzed using flow cytometry.The gene expression of p21,p53 and PCNA was evaluated by RT-PCR.Results The proliferation of SMMC-7721 cells was significantly inhibited in a dose-and time-dependent manner after UA treatment.Flow cytometric analysis showed SMMC-7721 cells treated with UA were accumulated in S phase in the cell cycle.The genes expression of p21 and p53 was increased in SMMC-772 cells cultured with UA,while the gene expression of PCNA was decreased.Conclusion UA has significant inhibition effect on the growth of SMMC-7721 cells.It can block the cell cycle in the S phase.Cell cycle arrest is related with the expressions of p21,p53 and PCNA.
