摘　　要：目的探讨南美响尾蛇神经毒素(crotoxin)对吉非替尼(iressa)诱导人肺鳞癌SK-MES-1细胞凋亡及其对吉非替尼的抗肿瘤效果的影响。方法应用MTT法和流式细胞仪annexinV-PI双标法研究crotoxin及其与iressa联用对人肺鳞癌SK-MES-1细胞促凋亡作用。结果 Crotoxin作用浓度大于或等于25μg/ml时对人肺鳞癌SK-MES-1细胞有明显生长抑制作用,与对照组相比差异有统计学意义(P<0.05),与iressa联合作用明显高于单用crotoxin和iressa,其Q值均在0.85～1.15之间,两者呈现出相加的抗肿瘤效果。结论 Crotoxin对人肺鳞癌SK-MES-1细胞的生长有抑制作用,并呈浓度依赖性,与iressa联合应用对人肺鳞癌SK-MES-1细胞有明显的促凋亡作用。Objective To investigate the effects of crotoxin on the apoptosis induced by iressa in SKMES -1 cells and its effect on the anti-tumor effect of iressa.Methods MTT was used measure the inhibitory effect of iressa and crotoxin on the growth of SK-MES-1 cells.The cell apoptosis was studied by annexin V-PI double labeling.Results Crotoxin inhibited the growth of SK-MES-1 cells when its concentration was larger than or equal to 25μg /ml,it was significantly different compared with that in the control group（P〈0.05）.And crotoxin strengthened the anti-tumor effect of iressa when it combined with iressa,suggesting the additive actions of the two drugs.Conclusion There are dose-effect relationships between crotoxin and the SK-MES-1 cells.And the apoptosis of SK-MES-1 cells can be increased significantly when it is combined with iressa.
