摘 要:目的探讨胞外信号调节激酶(ERK)和核因子-κB(NF-κB)在胃腺癌及癌旁正常组织中的表达及其与临床病理参数之间的关系。方法通过免疫组化及RT-PCR法,检测36例胃腺癌组织及癌旁正常组织中ERK和NF-κB的蛋白及mRNA表达情况,分析组织中蛋白的表达率及表达强度与临床病理参数之间的关系。结果 (1)胃腺癌组织中ERK、NF-κB蛋白及mRNA表达显著高于癌旁正常组织(均P<0.01);(2)ERK及NF-κB在低分化、Ⅲ-Ⅳ期的胃腺癌组织中显著升高(均P<0.01),ERK在有淋巴结转移的胃腺癌组织中表达升高(P<0.05),而NF-κB在胃腺癌组织中的表达与淋巴结转移无明显的相关性(P>0.05),ERK及NF-κB在胃腺癌组织中的表达与患者的性别、年龄、病变部位亦无相关性(均P>0.05);(3)ERK和NF-κB的表达之间呈正相关(r=0.653,P<0.01)。结论胃腺癌组织中ERK和NF-κB表达上调,两者的表达成正相关。两者在胃癌的发生发展中可能起重要作用,为临床早期诊断、预后判断及治疗胃癌提供了新的思路。Objective To detect the expression of extracellular signal-regulated kinase(ERK) and nuclear factor-kappa B(NF-κB ) in tissue of gastric adenocarcinoma and normal gastric tissue beside carcinoma, and to research the relationship between them and their clinical patho-parameters.Methods By the methods of immunohistochemistry and RT-PCR,the expression of protein and mRNAs of ERK and NF-κB in 36 cases of gastric adenocarcinoma and normal tissue beside carcinoma was detected and the relationship between the rate and intensity of its expression and clinical patho-parameter in the tissue was analyzed.Results(1 ) The expression of protein and mRNAs of ERK and NF-κB in gastric adenocarcinoma were higher than that in the normal gastric tissue beside carcinoma(P〈0.01).(2) The expression of ERK and NF-κB was significantly higher in the gastric adenocarcinoma in poorly differentiated andⅢ-Ⅳstage(P〈0.01).The expression of ERK was significantly higher in the gastric adenocarcinoma with aversion of conglobate gland(P0.05 ),while the NF-κB was opposite(P〈0.05 ).The expression of ERK and NF-κB had no link with sex,age and diseased region(P〉0.05).(3) Moreover the expression of ERK positive correlated with NF-κB in gastric adenocarcinoma(P〈0.01).Conclusion The expression of ERK and NF-κB is higher in gastric adenocarcinoma.There is positive correlation between them. They may take an important role in the development and the growth of gastric carcinomas and to supply new way in early diagnosis,judgement prognosis and treatment of gastric cancer.
