摘  要:目的研究跨损伤DNA合成通路中的关键基因RAD6、RAD18、PCNA和UBC13在人食管鳞癌与癌旁组织中的表达差异。方法采用SYBRGreen实时定量PCR技术检测60例食管鳞癌患者癌组织和其中的24例癌旁组织中RAD6、RAD18、PCNA和UBC13基因表达情况。结果 RAD6和UBC13在食管鳞癌与癌旁组织中的表达差异均无统计学意义(均P>0.05);RAD18和PCNA的表达在食管鳞癌中均显著上升,与癌旁组织相比差异均有高度统计学意义(均P<0.01)。结论 RAD18和PCNA的表达升高可能是食管鳞癌的一个特征。RAD18和PCNA表达的升高可能与食管鳞癌中跨损伤DNA合成途径作用增强有关。Objective To detect the expression differences of the key gene,including RAD6, RAD18,PCNA and UBC13,in translesion DNA synthesis between human esophageal squamous cell carcinoma specimens(ESCCs) and its adjacent tissue.Methods The expression levels of RAD6,RAD18, PCNA and UBC13 in 60 esophageal squamous cell carcinoma specimens and 24 adjacent tissues of esophageal squamous cell carcinoma were measured by SYBR Green real-time quantitative PCR.Results There was no significant expression difference RAD6 and UBC13 between human esophageal squamous cell carcinoma specimen and adjacent tissue of esophageal squamous cell carcinoma(both P〉0.05), while RAD18 and PCNA was significantly up-regulated in cancer tissues(P〈0.01).Conclusion The up-regulation of RAD18 and PCNA may be one of the characteristic of esophageal squamous cancer and may be associated with the strengthening of translesion DNA synthesis in esophageal squamous cell carcinoma.
