摘  要:目的 探讨P53及METTL-9基因在结肠的正常或炎症组织,癌组织及癌旁组织中的表达情况,研究其与结肠癌发生发展的关系.方法 将标本分为:正常或炎症组(5例正常和3例炎症结肠组织),癌组(22例)及癌旁组(22例).HE染色后在光学显微镜下观察组织细胞的形态学情况;应用实时荧光定量PCR方法 检测各组织中P53和METTL-9基因的表达情况.结果 正常或炎症组中P53基因的表达明显高于癌组和癌旁组(P<0.05),癌组中P53基因的表达明显低于癌旁组(P<0.05);正常或炎症组中METFL-9基因的表达明显低于癌组和癌旁组(P<0.05),癌组织中METTL-9基因的表达明显高于癌旁组织(P<0.05);癌症组织中P53基因与METTL-9基因呈负相关(P<0.05).结论 P53基因的异常表达在结肠癌发生过程中起着重要的作用;其下游基因METTL-9在结肠癌中高表达,转录翻译生成的蛋白酶可能导致结肠癌中DNA高度甲基化,从而促进结肠癌的发生发展.Objective To explore the expression of P53 and mettl-9 gene in colon cancer tissues and inflammation tissues and normal adjacent tissues and study its relationship with colon cancer.Methods The morphologic situation of histiocyte after H.E.stained was observed.The real-time fluorescent quantitative PCR was applied to detect the gene expression of P53 and METFL-9.Results The expression of P53 gene in inflammation tissues was significantly higher than that in cancer tissues and adjacent tissues(P 〈0.05).The expression of P53 gene in cancer tissues was significantly lower than adjacent tissues and infiammation tissues.METTL-9 gene expression in inflammation tissues was significantly lower than cancer tissues and adjacent tissues(P 〈0.05).In cancer tissues,P53 gene and METFL-9 gene were negative related.Conclusions The mutations of P53 gene played an important role in the course of colon cancer,its downstream gene METTL-9 was highly expressed in colon cancer,and the 9-methyl-converting enzyme which produced by transcription and translation might lead gene hypermethylation in the colon cancer,and thereby promoted the development of colon cancer.
