来曲唑治疗子宫内膜癌的实验研究

摘　　要：目的:利用ER+、PR+的人子宫内膜癌细胞株RL-952种植于BALB/c雌性裸鼠形成移植瘤模型后,通过观察移植瘤生长体积的变化、PTEN以及Ki-67的变化来研究来曲唑对子宫内膜癌的治疗效果。方法:将对数生长期的人子宫内膜癌RL-952细胞接种于裸鼠身上形成移植瘤模型,分别给予来曲唑、甲地孕酮、来曲唑+甲地孕酮及生理盐水来对比药物对移植瘤生长的影响。结果:来曲唑组给药后瘤体体积明显小于甲地孕酮组和空白对照组,但与来曲唑+甲地孕酮组差异无统计学意义。PTEN的表达率在来曲唑组、甲地孕酮组、来曲唑+甲地孕酮组均高于空白对照组,Ki-67在各组中的表达无差异。结论:来曲唑对子宫内膜癌移植瘤的抑制作用明显,且与甲地孕酮相比较,优于甲地孕酮。Objective：To research the curative effect of letrozole in treatment of endometrial carcinoma by observing the changes of growth size of transplantable tumor,the changes of PTEN and Ki67 after establishing rat models of transplantable tumor by planting human endometrial carcinoma cell strain RL-952（positive estrogen receptor and progestogen receptor） into BALB/c female nude rats.Methods：Human endometrial carcinoma RL-952 cells at logarithmic growth phase were planted into nude rats to establish rat models of transplantable tumor,the rates were treated with letrozole,megestrol,letrozole combined with megestrol and normal saline,respectively;the effects of different drugs on the growth of transplantable tumor were compared.Results：The decrease extent of size of transplantable tumor in letrozole group after treatment was significantly lower than those in megestrol group and blank control group,but there was no significant difference in decrease extent of size of transplantable tumor between letrozole group and letrozole combined with megestrol group.The expression rates of PTEN in letrozole group,megestrol group and letrozole combined with megestrol group were significantly higher than that in blank control group,but there was no significant difference in expression rate of Ki-67 among the four groups.Conclusion：Letrozole inhibits transplantable tumor of endometrial carcinoma obviously,and its curative effect is superior to that of megestrol.
