胶质瘤中GLUT-1与GLUT-3表达的实验研究

摘　　要：目的探讨葡萄糖转运蛋白-1(GLUT-1)和葡萄糖转运蛋白-3(GLUT-3)在人脑胶质瘤中的表达及与病理分级的关系。方法用W estern b lot和逆转录-聚合酶链反应(RT-PCR)的方法检测80例胶质瘤中GLUT-1和GLUT-3的表达情况。结果 GLUT-1蛋白在Ⅰ、Ⅱ、Ⅲ、Ⅳ级胶质瘤中的表达分别为0.09±0.02、0.20±0.02、0.41±0.09和0.63±0.07;GLUT-3蛋白在Ⅰ、Ⅱ、Ⅲ、Ⅳ级胶质瘤中的表达分别为0.10±0.02、0.23±0.07、0.62±0.05和0.88±0.06。各组间有显著差异(P<0.01)。GLUT-1mRNA在Ⅰ、Ⅱ、Ⅲ、Ⅳ级胶质瘤中的表达分别为0.08±0.03、0.19±0.02、0.41±0.06和0.66±0.08;GLUT-3 mRNA在Ⅰ、Ⅱ、Ⅲ、Ⅳ级胶质瘤中的表达分别为0.17±0.01、0.36±0.09、0.64±0.09和0.89±0.11。各组间有显著差异(P<0.01)。结论胶质瘤中GLUT-1和GLUT-3表达随病理级别增高而增加,提示GLUT-1和GLUT-3在胶质瘤的发生过程中起重要作用。Objective To study the expression of glucose transporter protein-1（GLUT-1） and glucose transporter protein-3（GLUT-3） in human glioma and their relation to pathological grade.Methods A total of 80 cases of glioma were examined for GLUT-1 and GLUT-3 expression by Western blot and reverse transcription-polymerase chain reaction（RT-PCR）.Results The expression of GLUT-1 protein in grade I,II,Ⅲ and Ⅳ glioma was 0.09±0.02,0.20±0.02,0.41±0.09 and 0.63±0.07,respectively.The expression of GLUT-3 protein in gradeⅠ,Ⅱ,Ⅲ and Ⅳ glioma was 0.10±0.02,0.23±0.07,0.62±0.05 and 0.88±0.06,respectively.There was significant difference between different groups for both GLUT-1 and GLUT-3（P 0.05）.The expression of GLUT-1 mRNA in grade I,II,Ⅲ and Ⅳ glioma was 0.08±0.03,0.19±0.02,0.41±0.06 and 0.66±0.08,respectively.The expression of GLUT-3 mRNA in the grade I,II,Ⅲ and Ⅳ glioma was 0.17±0.01,0.36±0.09,0.64±0.09 and 0.89±0.11,respectively.There was significant difference between different groups for both GLUT-1 and GLUT-3（P 〈0.05）.Conclusion The expressions of GLUT-1 and GLUT-3 correlate positively with the malignancy of glioma,which indicates GLUT-1 and GLUT-3 may play an important role in the tumorigenesis of gliomas.
