摘  要:目的:观察淫羊藿提取物IC163对K562细胞是否有分化诱导作用。方法:用不同浓度IC163作用于K562细胞6d,通过W-G染色观察细胞形态学变化、细胞内血红蛋白测定、细胞内血红蛋白联苯胺染色和流式细胞术检测膜CD71和CD235a分化抗原4个方面来评价IC163对K562细胞的诱导分化作用。结果:8μmol/L的IC163可明显上调K562细胞膜CD71和CD235a分化抗原的表达,升高细胞内Hb含量,在形态上有趋向红系分化的改变。结论:IC163有诱导K562细胞分化的作用。Objective:To determine the differentiation-inducing potentiality of IC163 on K562 cells.Method:Differentiation induction experiment was performed by culturing K562 cells with the presence of IC163(0-64 μmol/L) for 6 days.The changes of morphology,cytochemisrty and cytophyletic characteristic markers(CD71 and CD235a) of K562 cells were observed before and after the treatment with IC163,by the methods of wright's-Giemsa staining,benzidine staining and flowcytometry respectivly.Result:K562 cells treated with 8 μmol/L IC163 were found differentiated and morphologically changed to polychromatic normoblasts and orthrochromatic normoblasts.Treatment of K562 cells with IC163 increased hemoglobin content,benzidine-positive cells from(4.03±1.46)%(0 μmol/L) to(13.39±2.49)%(8 μmol/L) and up-regulated the expressions of CD71 and CD235a.Conclusion:IC163 can induce K562 cells differentiation.
