摘 要:目的探讨慢性强迫游泳应激对大鼠脑组织内鸟苷酸激酶相关蛋白表达的影响。方法将30只健康雄性Sprague-Dawley大鼠按体重随机分为对照组、游泳应激组、游泳应激+MK801组,每组10只,建立动物模型,将游泳应激组、游泳应激+MK801组大鼠置于(100X80×80)cm^3方型容器中进行强迫游泳,每次1只,每只每天游10min,持续14d,游泳应激+MK801组大鼠每天游泳之前皮下注射MK801(1g·kg·d^-1)。观察3组不动时间、体重增长情况、液体消耗量。采用免疫印迹方法测定慢性强迫游泳应激后各组大鼠海马、前额皮质、基底结的鸟苷酸激酶相关蛋白表达水平。结果经慢性强迫游泳应激后,与对照组及游泳应激+MK801组相比,游泳应激组体重增长量、糖水消耗量、糖水偏爱百分比均显著减少(P〈O.05)。游泳应激组大鼠在第14d强迫游泳中,不动时间较第1d显著增加(P〈0.05),而游泳应激+MK801组则无显著变化。游泳应激组海马鸟苷酸激酶相关蛋白表达量显著高于对照组及游泳应激4-MK801组(P〈O.01),前额皮质及基底结的鸟苷酸激酶相关蛋白量3组间差异无显著性。结论慢性强迫应激对大鼠的心理行为和生理成长均产生影响,鸟苷酸激酶相关蛋白可能是慢性应激反应的相关蛋白。Objective To explore the influence of chronic torced swimming on rat Drain CKAP expression. Methods Thirty healthy male Sprague-Dawley rats were randomly assigned to control, swimming stress and swimming plus MKS01 group (each n= 10) according to body weight, animal models established, one each time was forced to swim in (100XSOX80)cma square type container, swimed for 10 rain/day for 14 days continuously, before swimming lg/kg/d MKS01 were subcutaneously injected to rats of the swimming plus MKS01 group. Motionless, weight gain and fluid consumption were observed. GKAP exprssion levels in hippocampus, prefrontal cortex and basal ganglia were determined with the Western Blot. Results Compared with the control and swimming stress+MK801 group, the percentage of weight gain, sucrose consumption and sucrose partialism lowered more significantly after chronic forced swimming (P〈0.05). Motionless time during swimming significantly increased on the 14th day than the 1st in the swimming stress group (P^0.05), but there was no significant change in the swimming plus MKS01 group. GKAP exprssion levels in the Hippocampus were significantly higher in the swimming stress t^an in the control and swimming stress+MKS01 (P〈0.01), those in prefrontal cortex and basal ganglia had no significant differences among 3 groups. Conclusion Chronic forced stress takes effect on rat psycho-behavior and physio-growth, GKAP may be associated protein of chronic stress response.
