摘 要:目的探讨二至天癸颗粒对克罗米芬(CC)诱导排卵小鼠子宫内膜整合素β3表达的影响。方法 40只生育期排卵障碍模型小鼠分中药组及CC组两组,每组20只,中药组予二至天癸颗粒加CC,CC组单纯予CC,20只非模型鼠为空白组。观察三组胚泡着床率、子宫内膜形态学变化,并采用免疫组化方法测定子宫内膜整合素β3的表达。结果与CC组比较,中药组模型小鼠恢复动情周期数明显增多(P<0.05);与CC组比较,中药组孕4 d小鼠子宫内膜形态学改变较优,适于着床;与CC组比较,中药组孕4d小鼠子宫内膜腔上皮、腺上皮及间质三种组织中整合素β3的表达水平均较强,且均具有统计学意义;中药组与空白组小鼠比较,着床胚泡数、着床率无显著性差异(P>0.05);整合素β3的表达在腺上皮及间质积分无显著性差异(P>0.05),在腔上皮空白组表达较中药组强,且具有显著性差异(P<0.05)。三组小鼠整合素β3在腔上皮、腺上皮中的表达均强于在间质中的表达,具有统计学意义。结论二至天癸颗粒提高CC诱导排卵小鼠子宫内膜腔上皮及腺上皮细胞中整合素β3的表达。Objective: To explore the effect of Erzhi Tiangui Granules (ETG) on the expression of endometrlal beta3-integrin in mice of induced ovulation with clomifene citrate (CC). Methods: Forty Kunming female mice with ovulation failure were randomized into TCM group (treated with CC and ETG) and CC group (treated with CC only), while other 20 healthy mice of the same age were as normal controls. Their implantation rate on the fourth day of pregnancy (PD4) and the morphological changes of endometrium were observed, and the expression of endometrial beta3-integrin was detected by immunohistochemistry. Results: More mice had their oestrous cycle recovered in TCM group than in CC group (P〈0.05). The endometrial morphological changes on PD4 in TCM group were significantly more suitable for implantation(P〈0.05). The immunostaining scores of beta3-integrin in luminal epithelium, glandular epithelium and stroma of endometrium were all higher in TCM group than in CC group (P〈0.05). The number of implanted blastocysts, implantation rate, and the expression of beta3-integrin in glandular epithelium and stroma in TCM group were comparable to those in normal controls (P〉0.05), while expression of beta3- integrin in luminal epithelium was lower in TCM group than in normal controls (P〈0.05). The expres- sions of beta3-integrin in luminaland glandular epithelium were both higher than that in stroma in all three groups (P〈0.05). Conclusions: ETG can help to restore the oestrous cycle and improve the expression of endometrial beta3-integrin and implantation rate in the mouse with ovulation failure.
