摘 要:目的研究表没食子儿茶素没食子酸酯(epigallocatechin-3-gallate,EGCG)对鼻咽癌裸鼠移植瘤放疗的增敏效应及survivin和VEGF的表达及意义。方法体外培养低分化鼻咽癌CNE-2细胞,接种于20只雄性裸鼠皮下,待肿瘤结节约4~6 mm时,随机分为生理盐水组、EGCG组(30 mg/kg)、放疗组和EGCG联合放疗组。相应的处理因素治疗3周后,拉颈处死动物,剥离瘤体称重,计算肿瘤抑瘤率,HE染色观察组织病理学改变。流式细胞术检测细胞凋亡,一步法RT-PCR检测瘤体组织中survivin和VEGF的表达变化。结果与生理盐水组、EGCG组、放疗组相比,EGCG联合放疗组能显著的抑制肿瘤生长,能明显的促进细胞凋亡(P<0.01)。survivin和VEGF表达水平在EGCG联合放疗组最低(P<0.05)。结论 EGCG能增强CNE-2细胞裸鼠移植瘤对放疗的敏感性,其机制可能与抑制survivin和VEGF的表达水平有关。OBJECTIVE To study the expression and significance of survivin and VEGF mRNA in xenotransplanted nasopharyngeal carcinoma after treated by EGCG combined with radiation in nude mice.METHODS Nasopharyngeal carcinoma CNE-2 cells were cultured in vitro and injected subcutaneously into twenty male nude mice to establish human tumor xenografts.When the diameter of the tumors reached 4 to 6 mm,the twenty nude mice were randomly divided into four groups: control group,EGCG group,radiation group,EGCG combined with radiation group.Three weeks later,the mice were killed and the tumor weight was measured to calculate the tumor inhibition rate.HE staining was used to observe the pathological changes of the tumor.Apoptosis rate was detected with flow cytometry.One step RT-PCR was applied to detect the expression of survivin and VEGF.RESULTS EGCG combined with radiation group could apparently inhibit the tumor growth compared with control group.Apoptosis rate were significantly increased in the other three groups except the control group,especially in EGCG combined with radiation group.Furthermore,the expression level of Survivin and VEGF in EGCG combined with radiation group were the lowest among the four groups(P 0.01).CONCLUSIONEGCG can improve the CNE-2 xenografts sensitivity to radiotherapy.The inhibition of the expression of Survivin and VEGF mRNA may be the possible mechanism.
