摘 要:目的:研究灵芝多糖(GLP)对长期大强度运动小鼠巨噬细胞活性及功能的影响。方法:实验小鼠50只随机分为盐水+对照组、盐水+运动组、低剂量GLP+运动组、中剂量GLP+运动组、高剂量GLP+运动组,每组10只。3个GLP组每天分别按50 mg/kg/日GLP(低剂量)、100 mg/kg/日GLP(中剂量)、200 mg/kg/日GLP(高剂量)灌胃。盐水+对照组和盐水+运动组每天灌胃等体积生理盐水,共4周。对照组不运动,4个运动组小鼠每天进行1次45 min负重5%游泳训练,每周6次,为期4周。测定4周后各组小鼠巨噬细胞分泌的NO、IL-1β水平和细胞内GSH及吞噬率、吞噬指数。结果:4周运动后盐水+运动组小鼠腹腔巨噬细胞分泌的NO、IL-1β水平显著下降。盐水+运动组小鼠腹腔巨噬细胞吞噬率显著下降。盐水+运动组小鼠腹腔巨噬细胞吞噬指数呈现下降趋势。中剂量GLP+运动组小鼠腹腔巨噬细胞分泌的NO、IL-1β水平显著高于盐水+运动组,高剂量GLP+运动组显著高于盐水+运动组和低剂量GLP+运动组,中、高剂量GLP+运动组小鼠细胞内GSH水平显著低于盐水+运动组和低剂量GLP+运动组,随GLP剂量增加GSH逐渐下降。低、中、高剂量GLP+运动组小鼠腹腔巨噬细胞吞噬率显著高于盐水+运动组,随GLP剂量增加巨噬细胞吞噬率逐渐提高。中剂量GLP+运动组小鼠腹腔巨噬细胞吞噬指数显著高于盐水+运动组和低剂量GLP+运动组。结论:GLP对大强度运动小鼠腹腔巨噬细胞吞噬功能及分泌IL-1β、NO具有明显的促进作用。Objective To investigate the effects of ganoderma lucidum polysaccharide(GLP)on macrophages activity and functions in long-term high intensity exercise mice.Methods 50 experimental mice were randomly divided into 5 groups: saline water + control group(S+C),saline water + training group(S+T),low dose GLP + training group(LG+T),medium dose GLP + training group(MG+T),high dose GLP + training group(HG+T),10 mice in each group.Different dosage of GLP were supplemented in GLP groups,50mg/kg/day for low dose group,100mg/kg/day for medium dose group and 200mg/kg/dayfor high dose group.S+C group and S+T group were treated with same volume of saline water per day,for 4 week.Four weeks load swimming training program with 5% body weight were conducted in training groups,45mins per day,6 days per week.After 4 weeks,the macrophage secreting NO,IL-1β and intracellular GSH,phagocytic percentage,phagocytic index of mice in each group were measured.Results After 4 weeks exercise,the macrophage secreting NO,IL-1β of peritoneal cavity in S+T group decreased obviously.The macrophage phagocytic percentage of peritoneal cavity in S+T group decreased obviously.The macrophage phagocytic index in S+T group showed decrease.Macrophage secreting NO,IL-1β in MG + T was obviously higher than that in S+T group,Macrophage secreting NO,IL-1β in HG+T were obviously higher than that in S+T group and LG+T group.The GSH in medium and high dose GLP + T groups was obviously less than that in S+T and LG+T group,GSH decreased with the increase of GLP dosage.The macrophage phagocytic percentage of peritoneal cavity in low,medium and high dose GLP+T group were obviously higher than that in S+T group,the macrophage phagocytic percentage increased with increase of GLP dosage.Macrophage phagocytic index of peritoneal cavity in MG+T group was obviously higher than that in S+T and LG+T group.Conclusion GLP obviously promoted peritoneal macrophage phagocytic function and IL-1β
