摘  要:目的观察急性高眼压损伤后RhoA、Rho激酶-2(Rho-associated kinase-2,ROCK-2)和内皮素(endothelin-1,ET-1)在大鼠视网膜中的分布、表达及其相关性。方法 30只SD大鼠随机分为正常组(N组)、急性高眼压损伤后1d组(H1组)、3d组(H3组)、5d组(H5组)和7d组(H7组),制作急性高眼压模型后,用免疫组织化学法观察各组RhoA、ROCK-2和ET-1在视网膜中的分布、平均吸光度(average optical density,AOD)及其相关性,Western blotting法观察各组RhoA和ROCK-2在视网膜中的蛋白表达量。结果 N组神经节细胞层中仅见散在几个RGCs中表达RhoA、ROCK-2或ET-1,其余各层均未见相应的阳性染色;损伤组视网膜中RhoA、ROCK-2和ET-1的分布范围随损伤时间的延长而扩大,其AOD值均高于相应N组(H1和H3组:均为P<0.05;H5和H7组:均为P<0.01),其中ET-1表达量分别与RhoA和ROCK-2表达量呈显著正相关(r分别为0.58738和0.80667,均为P<0.05);H1、H3、H5、H7组视网膜中RhoA和ROCK-2蛋白表达均高于N组(均为P<0.05),并随时间的延长显著增加,两两比较差异均有统计学意义(均为P<0.05)。结论急性高眼压损伤可以激活视网膜中RhoA/ROCK-2通路,使RhoA和ROCK-2分布范围变广、表达增加,并诱导ET-1释放增加,二者之间有显著正相关性。RhoA/ROCK-2通路的激活在大鼠急性高眼压后视网膜损伤中起到重要作用。Objective To investigate the distribution and expression of RhoA,Rho-associated kinase-2(ROCK-2)and endothelin-1(ET-1)in rat retina after acute high intraocular pressure(IOP)and their correlation.Methods Thirty SD rats were divided into 5 groups at random:normal group(group N),1 day after acute high IOP group(H1 group),3 days group(H3 group),5 days group(H5 group),and 7 days group(H7 group).After the high IOP models were established,immunohistological assay was carried out on paraffin sections of retina and the distribution and average optical density(AOD)of RhoA,ROCK-2 and ET-1 were observed,meanwhile the correlation of ET-1 with RhoA and ROCK-2 were analyzed.The expressions of RhoA and ROCK-2 protein were analyzed by Western blotting.Results In group N,RhoA,ROCK-2 or ET-1 was only distributed in few cells of ganglion cells layer and not found in other layers.The distributions were expanded along with the extension of time after acute high IOP,their average optical density values were higher than group N correspondingly(P〈0.05 between group H1 and H3,P〈0.01 between group H5 and H7).Moreover,there was prominently positive correlation of the expression of ET-1 with one of RhoA or ROCK-2(r=0.587 38,0.806 67,both P〈0.05).The expressions of RhoA and ROCK-2 protein in group H1,H3,H5,H7 were obviously increased compared with that in group N(all P〈0.05),and it augmented along with the extension of time after acute elevated IOP.Furthermore,there were statistical differences between adjacent two groups(all P〈0.05).Conclusion RhoA/ROCK-2 passage way is activated by injury of acute high IOP,its distribution in retina is expanded and the expressions of RhoA,ROCK-2 and ET-1 are enhanced,and there is prominently positive correlation.The activation of RhoA/ROCK-2 passage way plays an important role in rat retinal injury after acute high IOP.
