摘 要:从江苏苏州某污水处理厂的活性污泥中初筛分离出21株产絮凝剂的微生物菌株,复筛得到1株具有较高絮凝活性的絮凝剂产生菌,将其命名为XNJ1。通过16S rRNA基因序列同源性分析并结合生理生化特征分析,将菌株XNJ1鉴定为居幼虫普罗威登斯菌属(Providencia sp.)。试验结果表明,菌株XNJ1对高岭土悬浮液的最佳絮凝生长条件为:碳源为葡萄糖,氮源为尿素,pH值7.0~8.0,温度30℃左右,培养时间48 h。在最佳絮凝条件下,2%絮凝剂投加量对4%的高岭土悬浮液的絮凝率达到82.4%。上述研究结果表明:菌株XNJ1具有较好的生物絮凝研究价值和应用前景。In this paper,21 bacterial strains capable of flocculating were isolated from the activated sludge of a sewage treatment plant in Suzhou,Jiangsu Province.Among them,one bacterial strain,designated as XNJ1,was selected for further study because it showed the highest flocculating activity.Based on the morphological,biochemical characteristics and comparative analysis of the 16S rRNA genes,strain XNJ1 was identified as Providencia sp.The results showed that the medium composition and other cultural conditions had significant effects on bioflocculant production.The optimum culture conditions of strain XNJ1 were as follows:carbon source was glucose,nitrogen source was urea,cultural temperature was 30 ℃,initial pH was 7.0-8.0,and incubation time was 48 h.The flocculation activity of strain XNJ1 on kaolin solution was about 82.4% after optimization of the culture conditions.The study showed that strain XNJ1 had great potential in industrial applications.
