肺癌患者TCR Vβ亚家族表达及记忆性T细胞表达的研究

摘　　要：目的探讨肺癌患者外周血T细胞及胸水肿瘤浸润性淋巴细胞(TIL)的T细胞受体(TCR)Vβ亚家族及记忆性T细胞的表达情况。方法分离健康人和肺癌患者的T细胞,采用流式方法检测Vβ24个亚家族及CD45RO和CD28的表达情况,统计分析Vβ24个亚家族及记忆性T细胞的特点。结果 43例肺癌患者外周血TCR Vβ亚家族主要高表达Vβ3(11/43)、Vβ9(12/43)、Vβ13.2(7/43);15例胸水TIL主要高表达Vβ3(5/15)、Vβ9(3/15)、Vβ21.3(3/15);肺癌组记忆性T细胞(29/35)多于健康对照组(4/35)(P<0.05)。结论肺癌患者TCR Vβ亚家族存在选择性表达的情况;肺癌患者以记忆性T细胞为主。推测这种高表达Vβ亚家族的T细胞可能部分是记忆性T细胞。Objective To analyze the expressions of T cells receptor （TCR） Vβsubfamily and memory T cells of peripheral blood T ceils and pleural effusion tumor infiltrating lymphocytes （TIL）in patients with lung cancer. Methods T cells were isolated from lung cancer patients and healthy individuals. Then the expressions of TCR Vbeta 24 subfamilies、 CD45RO and CD28 were detected using the flow eytometry method （FCM）. Results The TCR V^subfamily expression of peripheral T cells show higher Vβ3 （ 11/43 ） Vβ9 （ 12/43 ） 、Vβ313.2 （ 7/43 ） in 43 patients with lung cancer; pleural effusion TIL show higher Vβ3 （ 5/ 15） 、Vβ9（3/15）、Vβ21.3 （3/15）in 15 patients with lung cancer; the memory T cells expression of lung cancer patients （ 29/35 ） are more than healthy individuals （ 4/35 ） （ P 〈 O. 05 ）. Conclusion Selective expression exists in the TCR Vbeta subfamily of lung cancer patients, T cells are primary memory T lymphocytes in lung cancer patients. We can speculate that some T cells of high expression TCR Vbeta subfamily may be the memory T cells.
