摘  要:目的:探讨抑癌方对小鼠结肠癌移植瘤微血管生成、血管内皮生长因子(vascular endothelial growthfactor,VEGF)及缺氧诱导因子-1α(hypoxia inducible factor-1 alpha,HIF-1α)影响。方法:用CT26结肠癌细胞建立小鼠结肠癌移植瘤模型,分阴性对照组、中药高、中、低剂量组及化疗药组5组,分别灌服生理盐水、中药及腹腔注射氟脲嘧啶(5-FU)。比较各组之间瘤重;采用免疫组化染色方法和显微图像分析技术检测不同浓度抑癌方对各组小鼠移植瘤微血管(MV)及VEGF、HIF-1a表达的影响。结果:与阴性对照组的瘤重相比较,抑癌方各组及5-FU组(阳性对照药,以下称化疗组)均有统计学差异(P<0.05);但中药各组瘤重均高于化疗组(P<0.05)。免疫组化染色结果显示阴性对照组癌巢间MV、VEGF及HIF-1a表达高于中药组和化疗组(P<0.05)。结论:抑癌方对小鼠移植瘤有一定抑制作用;能够减少肠癌移植瘤中血管形成及VEGF及HIF-1a的表达。Objective:To reseasech the effects of Yiaifang(YAF),a traditional Chinese medicine prescription,on microvessels formation and VEGF,HIF-1α expression in mice bearing transplanted colon cancer.Methods:The model of transplanted colon cancer was induced by colon cancer cell CT26 in mice,all mice were randomly divided into normal saline(NS)treated group were fed with NS;while mice in the high,medium and low dose YAF treated groups and chemotherapy group.Mice in NS treated group were fed with NS;while mice in the high-,medium-,low-dose YAF treated group were intraperitoneally injected with 5-fluorouracil.Tumor weights were observed and comparaed among all group after the mice were executed using dislocation of cervical vertebra.The microvess(MV)and VEGF,HIF-1a expression in the tumor tissues of each group were assayed by using immunohistochemical staining,and Microscope image analysis.Results:Tumor weights in high-,medium-and low-dose YAF treated group and the chemotherapy group were significantly lower than those in NS treat group(P〈0.05).While tumor weights in high-,medium-,and low-dose YAF treaed groups were significantly higher than those in chemotherapy group(P〈0.05).The expression of microvess(MV)and VEGF,HIF-1a in all dose YAF treated groups and chemotherapy group were significantly decrearsed comparaed with those in NS treated group(P〈0.05).Conclusion:YAF can inhibit the development of mouse transplanted tumor and decrease the expression of microvess(MV)and VEGF,HIF-1a.
