白介素17在三硝基苯磺酸诱导结肠炎小鼠结肠内的表达及益生菌的作用

摘　　要：目的评价IL-17在TNBS(三硝基苯磺酸)诱导结肠炎小鼠结肠中的表达情况及益生菌对IL-17的影响。方法双歧三联活菌干预组(B IFICO组)与阴性对照组(TNBS组)小鼠自实验前5天起分别给予双歧三联活菌混悬液或PBS灌胃,实验当天及实验后第5天给予TNBS/乙醇溶液灌肠,实验第7天处死。处死后行组织学评分评价结肠炎症情况,并以免疫组化方法评价结肠内IL-17的表达情况。结果 B IFICO组小鼠结肠组织学评分低于TNBS组(2.11±0.78vs3.22±0.97,P<0.05)。IL-17在上皮细胞中表达程度评分,TNBS组明显高于正常对照组(8.00±0.82vs3.50±0.55,P<0.01),B IFICO组低于TNBS组(6.78±0.83vs8.00±0.82,P<005);固有层中IL-17阳性细胞计数,TNBS组明显高于正常对照组(10.74±2.78vs0.90±0.70,P<0.01),B IFICO组低于TNBS组(4.07±1.39vs10.74±2.78,P<0.01)。结论 TNBS诱导结肠炎的小鼠结肠内IL-17表达明显增加,双歧三联活菌可能通过抑制IL-17的表达减轻小鼠的实验性结肠炎。Objective To evaluate the expression of interleukin-17（IL-17） and the effect of the probiotic BIFICO in 2,4,6-trinitrobenzene sulfonic acid（TNBS） induced murine colitis.Methods Mice in BIFICO group and TNBS group were respectively gavaged BIFICO or PBS daily from 5 days before the experiment until sacrifice at D7.TNBS/ethanol were clystered on the first day and fifth day respectively.Colons were removed to score for histological inflammation after the mice were sacrificed.Expression of IL-17 in colon was determined by immunohistochemistry method.Results Colonic microscopic score was lower significantly in BIFICO group than that in TNBS group（2.11±0.78 vs 3.22±0.97,P0.05）.The expression of IL-17 in intestinal epithelium was significantly higher in TNBS group than that in normal control group（8.00±0.82 vs 3.50±0.55,P0.01）,and reduced in BIFICO group（6.78±0.83 vs 8.00±0.82,P0.05）.IL-17 positive cell counts in lamina propria was higher in TNBS group than that in control group（10.74±2.78 vs 0.90±0.70,P0.01）,and reduced in BIFICO group（4.07±1.39 vs 10.74±2.78,P0.01）.Conclusion Expression of IL-17 significantly increased in TNBS-induced colitis.BIFICO could alleviate the experimental colitis of mice by inhibiting IL-17 expression.
