摘  要:目的 研究药物 S R4233 对人脑胶质瘤细胞细胞毒作用及效应特点。方法 采用成克隆分析法分析乏氧及空气条件下 S R4233 及顺铂单独用药和联合用药在不同药物浓度及作用时间下药物毒性及作用特点。结果  S R4233 对乏氧的脑肿瘤细胞具有特异毒性,乏氧细胞毒性比( H C R) 值为47 .1 而作为阴性对照组的顺铂对乏氧细胞及有氧细胞的毒性作用相似,其 H C R 值为1 .0 。在乏氧条件下, S R4233 和顺铂联合用药对乏氧细胞的杀灭作用大于单独用药,其中先给 S R4233 的效果优于先给顺铂,这可能是由于 S R4233 首先杀灭了大部分的乏氧细胞而顺铂则杀灭了剩余的乏氧程度较轻的细胞,从而导致了给药顺序不同所致的效应差别。结论 实验结果显示: S R4233 是一种对乏氧细胞具有选择毒性的抗癌药,并对脑肿瘤的治疗具有一定应用潜力。Objective To investigate the effect of SR4233(a new bioreductive anticancer drug) to U251 cells (glioma cells). Methods Clonogenic assay was used to determine the effect of SR4233 on U251 cells. On the day of experiment, cells were given different drug dose on spaced several times apart under hypoxic or aerated condition. Results The results showed that SR4233 preferentially killed hypoxic cells,hypoxic cytotoxicity ratio (HCR) for SR4233 and cisplatin were 47.1 and 1.0 respectively under hypoxic condition. The cytotoxicity was stronger in combined SR4233 and cisplatin than SR4233 alone.The effect was better if SR4233 was given first than if cisplatin was given first. The reason of this sequential importance may have been that SR4233 killed most of the hypoxic cells and cisplatin annihilated the rest.Conclusions The results in our experiment suggest that SR4233 possess selective toxicity for hypoxic glioma cells. It is likely that SR4233 will be useful in the clinical practice of treating brain tumors.
