摘  要:评价生物降解性的环胞素 A 硬脂酸毫微球对兔眼实验性增殖性玻璃体视网膜病变的防治作用。方法: 成年健康灰兔 30 只, 随机分组, 对照组 20 只眼内注入 1% 空白毫微球悬液或磷酸缓冲液 ( P B S); 环胞素 A 毫微球组 (含环胞素 A08m g/m l或 16m g/m l, 2 组各20 只眼)。各组均先后2 次行气体压迫玻璃体手术。第二次玻璃体注气后第 7 天, 所有实验眼玻璃体腔内注入成纤维细胞悬液 2×105 /01m l, 再分别注入空白毫微球, P B S或载药毫微球悬液 01m l。以间接检眼镜观察牵引性视网膜脱离发生情况, 共四周。结果: 4 周末时对照组、环胞素 A 毫微球 08m g/m l组、环胞素 A 毫微球 16m g/m l组的牵引性视网膜脱离的发生率分别为 90% 、526% 和30% , 两个环胞素 A 毫微球组与对照组相比经统计学处理有显著性差异 ( P< 001)。结论: 一次性注入环胞素 A 生物降解性毫微球能够有效地减少牵引性视网膜脱离的发生率。To evaluate the possibility of biodegradable cyclosporine A stearic acid nanoparticles (CyA SA NP) for the treatment of proliferative vitreoretinopathy(PVR)in an experimental rabbit model.Methods:Thirty pigmented rabbits (59 eyes) were divided into three groups.The nanoparticles without drug(10 eyes) and phosphate buffer solution(PBS)(10 eyes)were injected intravitreally as the control.The CyA SA NP containing 0 8mg/ml (19 eyes) or 1 6mg/ml(20 eyes) of CyA were injected into other two groups respectively.Seven days after second gas compression vitrectomy,2×10 5 fibroblastes and CyA SA NP(or control)were injected in all eyes sequentially.The morbidity of tractional retinal detachment (TRD) were observed by ophthalmoscope for 4 weeks.Results:4 weeks later,the morbidity of TRD in control,0.8mg/ml and 1.6mg/ml CyA SA NP group was 90%,52 6%,and 30% respectively.Significant statistical differences were found between the control group and the two treated groups (P<0 01).Conclusions:The CyA SA NP containing 0 8mg/ml or 1 6mg/ml of CyA can inhibit the occurrence of TRD effectively.
