贵州苗族群体七个短串联重复序列基因座的遗传多态性

摘  要:目的调查贵州苗族群体D3S1358、D18S51、TH01、D21S11、PentaE、D13S317和TPOX 7个短串联重复序列(short tandem repeats,STRs)基因座的多态性,评价它们在法医鉴定的应用价值。方法应用PCR复合扩增-毛细管荧光电泳技术,对180名贵州苗族随机个体及60例家系进行检测,计算群体遗传学参数。结果 D3S1358、D18S51、TH01、D21S11、PentaE、D13S317和TPOX分别检出6、13、6、10、17、6、和5个等位基因;14、34、13、24、44、15和9个基因型;Mp、Dp、PIC、PE和He值分别为:0.160、0.044、0.014、0.063、0.030、0.113和0.275;0.840、0.956、0.862、0.937、0.970、0.887和0.725;0.640、0.830、0.650、0.800、0.900、0.720和0.520;0.445、0.751、0.366、0.773、0.779、0.699和0.412;0.711、0.878、0.658、0.889、0.892、0.852和0.689;基因型分布符合Hardy-Weinberg平衡定律;60例家系调查证实上述基因座均符合孟德尔常染色体共显性遗传,未发现突变。结论 D3S1358、D18S51、TH01、D21S11、PentaE、D13S317和TPOX基因座在贵州苗族群体中具有较高的多态性。可以作为该群体法医学亲子鉴定和个人识别的遗传标记。Objective:To investigate the polymorphisms of the seven short tandem repeat(STR) loci: D3S1358,D18S51,TH01,D21S11,PentaE,D13S317 and TPOX of Chinese Hmong population in Guizhou,and evaluate the forensic science application.Methods: PCR and ABI-310 Fluorescent Capillary Electrophoresis techniques were used to analyze the DNA samples from unrelated individuals of Chinese Hmong ethnic group in Guizhou.Results: D3S1358,D18S51,TH01,D21S11,PentaE,D13S317 and TPOX showed separately: 6,13,6,10,17,6 and 5 alleles;14,34,13,24,44,15 and 9 genotypes;Mp,Dp,PIC,PE and He showed seperately:0.160,0.044,0.014,0.063,0.030,0.113 and 0.275;0.840,0.956,0.862,0.937,0.970,0.887 and 0.725;0.640,0.830,0.650,0.800,0.900,0.720 and 0.520;0.445,0.751,0.366,0.773,0.779,0.699 and 0.412;0.711,0.878,0.658,0.889,0.892,0.852 and 0.689.The genotype distributions of the seven STR loci showed no deviation from the Hardy-Weinberg equilibrium.The parentage testing of 60 cases revealed an autosomal codominant inheritances and no mutations happened to seven STR loci.Conclusion: These data indicate that D3S1358,D18S51,TH01,D21S11,PentaE,D13S317 and TPOX have good polymorphisms,with high probability of exclusion and probability of discrimination power as well as being loci available as the genetic markers to forensic parentage testing and personal identification in Hmong population.
