摘  要:目的探讨激光捕获显微切割技术(LCM)联合短串联重复序列(STR)多态性分析用于葡萄胎受精类型鉴别的可行性。方法首先通过LCM获得葡萄胎病病理组织的滋养细胞,再用16个位点(D8S1179,D21S11,D2S1338,CSF1PO,TH01,TPOX,D3S1358,D13S317,D16S319,D19S433,FGA,D18S51,D5S818,VWA,D7S820,Am)复合微卫星序列-PCR方法对61例病理学诊断为葡萄胎标本进行分析,明确其受精类型。结果用LCM技术从61例标本中获得滋养细胞后,成功提取DNA46例,浓度范围为:6.2-114.5(ngμ/l)。经STR多态性分析确定空卵单精子受精完全性葡萄胎13例,空卵双精子受精完全性葡萄胎13例,部分性葡萄胎20例。结论 LCM联合多重STR多态性分析能够确定葡萄胎的受精类型。Objective:To investigate the feasibility of short tandem repeats(STR) polymorphisms with Laser capture micro-dissection(LCM) in determination of fertilization types of hydatidiform mole(HM).Methods: The pure trophoblastic cells of HM were obtained by LCM and all the specimens were examined by multiplex STR—PCR(16loci: D8S1179,D21S11,D2S1338,CSF1PO,TH01,TPOX,D3S1358,D13S317,D16S319,D19S433,FGA,D18S51,D5S818,VWA,D7S820,Am),then to determine the fertilization types of HM.Results: the numbers that we successfully extracted DNA from all the 61 cases by LCM were 46 cases,the range of DNA's concentration: 6.2-114.5(ng/μl).And through the analysis of STR-PCR we found that complete hydatidiform moles(CHM) from fertilization of an anuclear empty ovum by a haploid sperm were 13 cases,while complete hydatidiform moles(CHM) from dispermic fertilization were 13 cases,and the remain 20 cases were partial hydatidiform moles(PHM).Conclusion: The results suggested that short tandem repeats(STR) polymorphisms with laser capture micro-dissection can determine the fertilization types of HM.
