摘 要:目的 研究大鼠蛛网膜下腔出血(subarachnoid hemorrhage,SAH)后自噬的激活情况及脑水肿的时程变化,探讨自噬这一细胞行为在SAH后早期脑损伤中的作用,以期为SAH的治疗提供新策略和新靶点.方法 采用颈内动脉刺破法制备大鼠SAH模型,采用数字表法随机分为正常对照组和SAH 6h组、24h组、72h组,通过脑干湿重法测定SAH早期脑水肿的程度,并通过免疫印迹技术检测各组脑组织中自噬相关蛋白LC3的表达.结果 脑干湿重法测脑含水量显示SAH模型建立后6h出现脑含水量增多,至3d时明显升高(P<0.05).Western blot结果显示:SAH后24h LC3 II/LC3 I较正常对照组即有明显增加(P<0.05),持续至72h仍有较高表达.结论 SAH早期大鼠大脑皮层组织神经元自噬被激活的同时早期脑损伤加重,与其表达时程具有相关性,大鼠SAH后早期自噬的激活可能加重了脑损伤.Objective To investigate the autophagy after Subarachnoid hemorrhage (SAH) in rats. Methods SAH model was induced by puncture of internal carotid artery in rats. Twenty four Sprague-Dawley rats were divided into 4 groups: normal group, SAH 6h, SAH 24h and SAH 72h groups with 6 rats in each. Dry-wet weight method was used to examine the degree of brain edema in SAH and Western-blot was performed to detect the expression of autophagy-related protein LC3. Results Dry-wet method showed that the water content of brain started to increase at 6h, and markedly increased at 72h after SAH. Western blot showed that expression of protein LC3 was increased at 24h after SAH,and remained positive at 72h after SAH. Conclusion Autophagy is activated after SAH in rats, which might be associated with the aggravation of SAH-induced cerebral injury.
