摘  要:目的:探讨CD133抗原在人结肠癌细胞SW620中表达是否具有可塑性。方法:对SW620细胞进行流式细胞活化荧光分析,分选出CD133-和CD133+细胞亚群。对这2种亚群细胞进行单层培养传代、悬吊法三维球培养及无糖或无血清培养液培养后,应用免疫染色与荧光定量PCR法检测CD133表达变化。结果:结肠癌SW620细胞中存在CD133-与CD133+两种亚群。分选后的CD133-与CD133+亚群细胞在单层培养时CD133表达无明显变化;在三维球培养时,CD133-亚群细胞中出现明显的CD133抗原及mRNA表达(P<0.05);而在无血清培养时,CD133-与CD133+两亚群细胞的CD133表达无变化;在无糖培养时,CD133-亚群细胞中出现CD133阳性表达。结论:人结肠癌SW620细胞中表面抗原CD133表达具有可塑性,可受不同培养条件的调节。Objective:To investigate the plasticity of CD133 antigen expression in human colon cancer cell line SW620.Methods:The CD133-and CD133+ cell populations were sorted from SW620 cells by using fluorescence activated cell sorting(FACS) method.The changes of CD133 expression were detected under the culture conditions of monolayer,hanging drop spheroid and glucose or serum withdrawal culture in CD133-and CD133+ subpopulations by using immuno-staining and fluorescence quantitative PCR.Results:There were two subpopulations including CD133-and CD133+ cells in human colon cancer cell lines SW620.There was no difference in the expression level of CD133 between these two subpopulations in monolayer culture,whereas the CD133 antigen and its mRNA expressions were induced in CD133-cell subpopulation in three-dimension spheroid culture(P0.05).The CD133 expression level was not changed in these two cell subpopulations in serum withdrawal culture,and the expression of CD133 antigen was induced in CD133-cell subpopulation in glucose withdrawal culture.Conclusion:The expression of surface antigen CD133 in human colon cancer cell lines SW620 can be modulated plastically in various cell culture conditions.
