摘 要:目的:制备和研究一种新的体外三维环境下肝细胞黏附培养的微载体支架-壳聚糖球形多孔微载体。方法:利用壳聚糖为基本材料,以2%乙酸为溶剂,采用液氮冷冻干燥技术制备壳聚糖球形多孔微载体,采用光学显微镜、扫描电子显微镜等仪器和方法对其形态结构及理化性质进行表征。结果:本研究成功地制备出壳聚糖球形多孔微载体,成球率>90%,内外多孔且相互连通,孔径在20~80μm之间,具有一定的密度、弹性、溶胀性、亲和性和形态结构,并具有良好的机械强度、稳定性和较大的比表面积。结论:本研究制备的壳聚糖球形多孔微载体为以其作为支架来进行肝细胞体外三维环境下的培养提供了一定的理论和实验依据。Objective:To prepare and study a new microcarrier support named spherical porous chitosan microcarrier,for hepatocyte adhesion culture in three-dimensional environment in vitro.Methods:The microcarrier with chitosan as the material and 2% peracetic acid as solvent was prepared by the liquid nitrogen freeze-drying technology.The optical microscope,scanning electron microscopy and other methods were used to characterize its morphous,physical and chemical properties.Results:The spherical porous chitosan microcarrier was successfully prepared in the project.Further more,the ball-rate was more than 90%.The porosity was occupied inside and outside of the microcarrier and interconnected.The pore size ranged from 20μm to 80μm..The microcarrier not only has a certain degree of density,elasticity,swelling,and affinity and morphology structure.but also has good mechanical strength,stability,and larger specific surface area.Conclusion: Spherical porous chitosan microcarrier provides a theoretical basis and experimental evidence for hepatocyte adhesion culture in three-dimensional environment of vitro as a support.
