摘  要:目的探讨血管紧张素Ⅱ-1型(AT1)受体在NAFLD发病机制中的作用。方法雄性SD大鼠32只随机分组,正常对照组、模型组各16只,适应性喂养1周后,模型组给予高脂饮食,正常对照组给予正常饮食,10周、14周后分两次平均处死各组8只大鼠,全自动生化分析仪测定肝功能、血脂,放射免疫计数器测定血管紧张素、醛固酮,运用免疫组织化学方法观察肝组织中AT1受体的表达。结果 10周末,模型组大鼠体质量增长高于正常对照组,14周末,模型组大鼠体质量增长低于正常对照组;10周、14周末模型组肝湿重、血清丙氨酸氨基转移酶(ALT)、总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL)、血管紧张素(AⅡ)浓度值均高于正常对照组;模型组大鼠高密度脂蛋白(HDL)均低于正常对照组;10周末时模型组AⅠ、AⅡ均较正常对照组升高,AⅠ在两组差异不具有统计学意义(P>0.05),14周末时模型组AⅠ、AⅡ均较正常对照组升高,差异具有统计学意义;模型组ALD均较正常对照组降低,差异不具有统计学意义;模型组有AT1受体表达,且14周时表达量高于10周时,正常对照组几乎不表达。结论 AT1受体在非酒精性脂肪性肝病大鼠肝组织中表达,且随着脂肪肝程度的加重表达量有所增加。Objective To investigate the role of Angiotensin II type 1 receptor in NAFLD Pathogenesis.Methods After one week′s adaptive feeding,32 male rats were randomly divided into control group (ordinary diet)and model group(high-fat diet)equally. In each group, 8 rats were killed at the end of the tenth week and the fourteenth week respectively. Liver function and blood lipid were monitored by automatic biochemistry analyzer. Angiotonin(AⅡ)and aldosterone(ALD) were measured by radioimmunoassay counter. The expression of AT1R in liver tissue were detected by immunohistochemistry. Results The rise of body weight in model group was greater than that in the normal control group at the end of the tenth week,while it was less at the end of the fourteenth week, liver weight, serum ALT, TC, TG, LDL and AⅡ in model group were higher than those in normal control group,while the HDL was significantly lower at the end of the tenth and fourteenth week. At the end of the tenth week, Serum angiotonin 1 and 2 in model group were higher than those in control group, but AI showed no statistical significance (P0.05); At the end of the 14 th weeks, Serum angiotonin 1 and 2 in model group were significantly higher than those in control group. Serum ALD in model group was lower than that in normal control group without statistical significance; the expression of AT1R existed in model group,with higher expression level at the end of the 14th week; there was nearly no expression in control group.Conclusion Angiotensin II type 1 receptor was expressed in Nonalcoholic fatty live rats, and the expression level increased gradually with the aggravation of the fatty liver lever.
