摘 要:目的研究神经干细胞移植对β淀粉样蛋白(Aβ)所致痴呆大鼠记忆功能及海马组织丙二1-40醛(MDA)、超氧化物岐化酶(SOD)的影响。方法取新生大鼠脑皮质进行神经干细胞的分离、培养、鉴定,传代培养2-3代;选取30只健康SD大鼠随机分为3组,10只/组。对照组:侧脑室注入蒸馏水5μl;模型组:侧脑室注入Aβ5μl;干细胞移植组:侧脑室注入Aβ5μl干细胞悬液2μl,4周后检测各组大鼠水迷宫潜伏1-401-40期及海马组织MDA和SOD水平。结果与对照组比较,模型组+大鼠水迷宫潜伏期明显延长(P<0.01),海马组织MDA和SOD水平明显升高(P<0.01)。与模型组比较,干细胞移植组大鼠水迷宫潜伏期明显缩短(P<0.05),海马组织MDA水平明显下降(P<0.05),SOD活性没有明显差异(P>0.05)。结论神经干细胞移植可改善Aβ1-40所致痴呆大鼠的记忆功能,降低海马组织氧化应激水平。Objective To study the effect of neural stem cell transplantation on cognitive function, and the levels ofmalondialdehyde (MDA) and superoxide dismutase (SOD) in hippocampus of dementia rat reduced by amyloid beta (A β1-40)- Methods Neural stem cells (NSC) obtained from the cerebral cortex of neonatal rats were separated, cultured, labeled and passaged for 2-3 generations. 30 SD rats were random divided into three groups (10 each group): control group(5 μl distilled water injected into lateral ventricular), model group(5μl βμ 1-40 injected into lateral ventricular), neuron stem cells transpantaiion(NSC) group(5 μ1 Aβ1-40 and 2 txl of NSC suspension injected into lateral ventricular). The latent period was tested by Morris water maze, and the levels of MDA and SOD in hippocampal tissue were determined. Results The latent period was significantly longer by Morris water test (P〈0.01), the levels of MDA and SOD in hippoeampal tissue were significantly higher in model group than in control (P〈0.05). The latent period was significantly shorter and MDA level was significantly decreased in NSC group than in model group, hut with no significant difference for SOD activities between NSC group and model group (P〉0.05). Conclusion The improvement of NSC transplantation on cognitive function of dementia rat induced by A β1-40might be related to the decrease of oxidative stress levels in hippocampal tissue.
